GENE THERAPY AND MOLECUALR GENETICS OF
HEMATOPOIETIC STEM CELLS

Guy Sauvageau, M.D., Ph.D., FRCP(C)

Institut de Recherche Clinique de Montreal (IRCM)

Montreal, Quebec, Canada

Project description:

(description en francais)

Our laboratory explores the hypothesis that transcription factors which control proliferation and
differentiation of embryonic tissues may also regulate these processes in normal and possibly abnormal
(e.g. leukemic, etc.) hematopoiesis. The following objectives are being persued in our laboratory:

1.to study the function of selected transcription factors whose expression is restricted to primitive
     hematopoietic stem cells,
2.to use these genes in order to manipulate the behaviour of hematopoietic stem cells and,
3.to identify and develop novel strategies to optimize gene transfer to these cells.

These goals are being addressed by using different experimental strategies such as experimental bone
marrow traplantation procedures (murine model), retroviral gene transfer, generation of cDNA library
starting from a single cell, in vitro differentioation of embryonic stem (ES) cells and generation of
transgenic and knock mice.

GENETIQUE MOLEDULAIRE DES CELLULES SOUCHES
HEMOPOIETIQUES ET THERAPIE GENIQUE

Guy Sauvageau, M.D., Ph.D., FRCP(C)

Postdoctoral Positions in Neuroscience
Two positions available to study the neuronal cytoskeleton and
neuronal morphogenesis. Our work attempts to define the mechanisms
that establish the cytoskeleton during axon growth. Toward this
end, we combine microinjection of tagged cytoskeletal proteins and
digital image analysis to examine the transport and assembly of
cytoskeletal elements in growing neurons. A second project examines
functional specializations of cytoskeletal proteins in axonal and
dendritic growth using antibody injection approaches. Additional

Postdoctoral fellowship: Electrophysiology of central respiratory chemoreception.
Program in Neuroscience
Departments of Neurology, and Cellular & Molecular Physiology
Yale University School of Medicine

Applications are invited for a Postdoctoral Fellowship Position to study the
effect of pH / CO2 on neuronal electrophysiology. Previous work in this
laboratory has identified neurons in the rostral ventral medulla with a
uniquely highsensitivity to respiratory acidosis that are candidates for
central respiratory chemoreceptors (J. Neurophysiol., 73(3):933-944, March,
1995). This finding provides a unique opportunity to rigorously define the
mechanisms of central respiratory chemoreception. The candidate
will use whole-cell, perforated-patch, and single-channel patch-clamp
recordings from neurons in thin brain slices and in culture to define
the mechanisms of transduction of respiratory acidosis into changes in
neuronal excitability.

Techniques currently employed in my laboratory or in collaboration
with other laboratories to address the function of these neurons include:

- Whole cell, perforated patch and single channel
		patch clamp recordings.
- Thin brain slices and cultured neurons.
-	Biocytin and HRP injections into neurons.
-	Light microscopic and electron microscopic
		analysis of neuroanatomy.
-	Rhodamine bead injections in vivo prior to
		in vitro electrophysiology experiments
		to determine neuronal projections.
-	Intracellular calcium and pH imaging.
-	mRNA analysis of neurotransmitters using in situ hybridization,
		and northern blotting.
-	Immunohistochemistry of neurotransmitters.

Applicants should have experience in electrophysiologic techniques.
Ph.D. in Physiology, Neuroscience, or Biology preferred.
Salary will be commensurate with experience.

Please send:
*   Curriculum vitae
* Brief statement of research interests
*   Reprints of previous work
*   Statement of citizenship
*   Address and telephone #

The Research Environment For Structural Molecular Biology At Leeds

The School of Biological Sciences at the University of Leeds is very large and
varied, and carries out high quality research in fields ranging from biophysics
to "whole organism" biology. The Departments of Biochemistry & Molecular
Biology (BMB) and Biology occupy contiguous space in linked buildings. BMB
provides the focus for the interdisciplinary Leeds Centre for Molecular
Recognition in Biological Systems (LCMRBS), funded by BBSRC, that additionally
encompasses elements of the School of Chemistry.

A very powerful research programme in structural molecular biology is well
established within LCMRBS, comprising groups working in X-ray crystallography
(group leaders: Prof. SEV Phillips, Prof. ACT North, Dr. AJ Geddes, Dr. MR
Parsons, Dr. J Jaeger [BMB]), NMR spectroscopy (Dr. JRP Arnold [Biology], Dr. J
Fisher [Chemistry]), electron crystallography (Dr. AKH Holzenburg [BMB]) and
protein folding and design (Dr. SE Radford and Dr. A Berry [BMB]). These groups
are well integrated in terms of exchange of personnel and collaboration on
projects of common interest. The X-ray equipment, NMR spectrometer and electron
microscopes are all grouped together in the same building, facilitating this
level of integration. The protein crystallography will move to a new, purpose-
built Biomolecular Sciences research building, contiguous with the existing
building, at the end of 1996. BMB is also the base for the Leeds sequence
database (OWL), which is still under active development, especially in the area
of motif recognition. Major areas of active structural molecular biology
research include protein-nucleic acid interactions (DNA and RNA), nucleic acid
structure, membrane proteins, copper-containing oxidases, ligand binding
proteins of the lipocalin family, and drug binding to DHFR.

The crystallographic group is equipped to international standards, with two
rotating-anode X- ray generators, a Siemens/Xentronics area detector system, a
Rigaku Raxis-II X-ray image plate system, a crystal cryocooling system, a
four-circle diffractometer and several precession cameras. NMR studies are
supported by a Varian Unity 500 NMR spectrometer, with associated computer
workstation and data processing facilities. The electron microscope suite
comprises a number of instruments, including a Philips CM10 for high resolution
studies. Computing facilities within BMB are very powerful, with a cluster of
five Alpha-AXP and six VAX workstations and servers, 4 Evans and Sutherland
PS300 graphics systems, and a number of Silicon Graphics workstations (12 in
BMB itself, and a further 18 owned by the University but sited in BMB and
available for our use).

2) What to do about "spams," i.e., junk mail, ads, etc.
-------------------------------------------------------
BIOSCI is a set of parallel USENET newsgroups (the "bionet" groups),
mailing lists, and a hypermail archive at URL http://www.bio.net/.
The same postings are distributed on all media (except for a small
number of mailing-list-only groups at net.bio.net). Unfortunately it
is becoming a despicable practice on the Internet (by a few people out
to make a fast buck) to do automated mass postings to thousands of
newsgroups and mailing lists. These attempts to grab free advertising
are refered to as "spams" in the usual, somewhat boneheaded, net
terminology. USENET is more susceptible to this practice, and many
spams originate on the USENET groups and then are passed on to the
mailing lists. However, spammers also get lists of mailing addresses
and hit these too, so neither medium is immune.

What should you do personally if you get junk mail?
---------------------------------------------------
Just delete it and move on without reading it further. Filing a
protest is becoming increasingly useless because spammers are often
disguising the addresses where the messages are sent from. Unless you
really understand Internet mail systems, your attempt at protest by
sending replies to the message will often end up being sent to the
address of an innocent person that the spammer is victimizing.

What can BIOSCI/bionet do to protect its newsgroups?
----------------------------------------------------
The only solution currently available is to moderate the newsgroup.
If this newsgroup is already moderated, then you are in good shape.
Moderation protects the USENET distribution from about 95% of the
spams that are being sent to date and protects the mailing lists
completely. Moderation means, however, that someone has to take the
time to review each message before it goes out. We have set up
software here that simply allows the moderator to forward to an
address at net.bio.net messages that (s)he wishes to have distributed.
This takes no more time than that needed to read the message and pass
it on, say about 1 min. per message.

Most newsgroups currently have a discussion leader who is responsible
for their newsgroup. The discussions leaders and their e-mail
addresses are listed in the BIOSCI Information Sheet which is
available on the Web at http://www.bio.net/. If a newsgroup is being
hit with too many junk postings, please contact the discussion leader
for that group and see if there is interest in moderating the group.
Please do not assume that by simply posting a complaint to the
newsgroup itself, anyone on the BIOSCI staff will act on your
complaint. With close to 100 newsgroups to run, the BIOSCI staff has
to rely on the discussion leaders of each newsgroup to report problems
directly to us at biosci-help@net.bio.net.

We will moderate any of our newsgroups if the discussion leader tells
us that the readership of the group wishes to do so and if a moderator
is willing to do the work. For most BIOSCI/bionet groups, this
entails only a few minutes of work each day.

Moderating a newsgroup will resolve probably 95% of the junk postings
on the USENET distribution. Unfortunately there are easy ways for
determined spammers to override the moderation mechanism on USENET,
but we can protect our e-mail subscribers from unwanted postings if
the newsgroup is moderated. You can also access our newsgroups over
the WWW at URL http://www.bio.net. While this Web interface will not
stop spammers from trying to post to the groups, this will give you
yet another way, besides using USENET news, to keep the junk out of
your personal mail files. For those of you with local USENET news
systems, the Web interface will also give you faster access to new
newsgroups and recent postings.

3) Examples of subscribing and unsubscribing to the mailing lists.
------------------------------------------------------------------
PLEASE NOTE: The BIOSCI management does NOT act on
subscription/unsubscription requests that are posted improperly to the
newsgroups and mailing lists. People who do this only bother everyone
on the lists to no avail. Please be sure to follow the proper
procedures below.

Gory details are in the BIOSCI Information sheets on the Web at
http://www.bio.net. Below we give an example utilizing the
METHODS-AND-REAGENTS list at both of our two BIOSCI sites:

Users in the Americas and Pacific Rim countries who use the BIOSCI
------------------------------------------------------------------
node at computer net.bio.net:
----------------------------

A) Determine the "listname" which is the <=8 character mail address
                                         ^^^^^^^^^^^^^
for the group. These can be found in the BIOSCI Info. Sheet. For
the METHODS-AND-REAGENTS group the mailing address is
methods@net.bio.net. The listname is the portion of the address to
the left of the @ sign, i.e., "methods". The listname is used with
the "subscribe" and "unsubscribe" commands illustrated below.

B) Mail all commands in the body of a mail message addressed to
biosci-server@net.bio.net. Do NOT send commands to the newsgroup
posting addresses! Leave the Subject: line blank, any text on it
   will be ignored.

C) In the body of your message put one or more of the following
commands with an "end" command on the last line, e.g.,

   subscribe methods
   unsubscribe methods
   end

Do NOT put your e-mail address or other text on these lines. The
server only allows you to cancel your subscription if the address
on your mail header matches the address on our mailing list.
Please ask for help at biosci-help@net.bio.net if your address has
changed, e.g., if you know you are on the list but the server tells
you that you are not a member.

Users in Europe, Africa, and Central Asia who use the BIOSCI node at
--------------------------------------------------------------------
computer daresbury.ac.uk (also known as dl.ac.uk):
-------------------------------------------------

To subscribe and unsubscribe to/from the BIOSCI lists, you need to
specify the full USENET newsgroup name with "bionet-news." prepended.
The USENET newsgroup names are listed in the BIOSCI Information sheet
on the Web at http://www.bio.net/. For the METHODS-AND-REAGENTS list
the USENET newsgroup name is bionet.molbio.methds-reagnts, thus the
appropriate commands are

    sub bionet-news.bionet.molbio.methds-reagnts

    unsub bionet-news.bionet.molbio.methds-reagnts

These commands are included in a message addressed to mxt@dl.ac.uk,
NOT to the newsgroup mailing addresses. As usual, include the text in
the body of the message as text on the Subject: line is ignored.

To unsubscribe from all the lists at the UK node, use

    unsub bionet-news

Please note that if the address in the list is different than the one
in your mail message header, you will not be able to unsubscribe by
this method. If you have problems, please mail biosci@daresbury.ac.uk.

4) The BIOSCI user address and research interest directory.
-----------------------------------------------------------
Please take this opportunity to add your name, address, and research
interest information to the BIOSCI User Address Database if you have
not already done so.

You can fill out the address form directly through our Web page at URL
http://www.bio.net/adrform.html.

The address database is reindexed nightly for WWW access (the URL is
http://www.bio.net/). If you are not directly on the Internet but can
reach it by e-mail, please use our waismail server to access the user
directory. waismail use is described above. You can also request a
user address form by e-mail from biosci-help@net.bio.net.

Please check your database entry from time-to-time to see if your
address information is still up-to-date. Because of our limited
personnel resources, we ask that you resubmit a *complete* form to
revise your entry; we only replace complete entries and do not have
resources to edit old forms.

				Sincerely,

				Dave Kristofferson
				BIOSCI/bionet Manager

Post-doctoral positions are available to study the molecular
mechanisms of gene regulation and development using Drosophila and
yeast as model systems. There are currently two major areas of
research in the laboratory.

1) Understanding how the Drosophila morphogenetic protein Bicoid
activates target gene expression in an all-or-none fashion and how it
controls embryonic pattern formation [see Driever et al. (1989) Nature
342, 149-154; Lehming et al. (1994) Nature 371, 175-179; Ma et al.
(1996) Development 122, vol.4, in press; Yuan et al., MCB,
submitted].

2) Understanding how transcriptional activators stimulate gene
expression and, in particular, how they communicate with the general
transcription machinery in living cells [see Ma and Ptashne (1987)
Cell 51, 113-119; Ma and Ptashne (1988) Cell 55, 443-446; Ruden et
al. (1991) Nature 350, 250-252; Ma (1992) Gene Expression 2, 139-146;
Shaw et al. (1996) MCB, submitted].

Interested candidates should have a recent Ph.D. and previous
research experience in at least two of the following areas: protein
chemistry, molecular biology, Drosophila genetics or yeast genetics.

Newsgroup Charter and Moderation Policy:

EMPLOYMENT/bionet.jobs.offered is to be used for the posting of job
openings in the biological sciences or professional level jobs that
support the work of biological scientists (such as for
computer/systems programming/support). Jobs in both the non-profit
and for-profit sectors may be posted without format restrictions.
For-profit jobs, however, must be for professional level positions
that support biology research or that require the hiring of someone
with biological expertise, i.e., routine administrative jobs in
commercial companies, e.g., secretarial positions at biotech
companies, will not be posted in this forum and should instead be sent
to misc.jobs.offered.

PLEASE BE SURE to use descriptive Subject: lines in your job postings.
It also helps to start with the abbreviated job location using the
country code and possibly the state/province, e.g.,

Subject: US/CA Postdoctoral postion in XXX

indicating that the job is in the United States/California. Subject
lines like "Job Opening" do not attract readers to your ad!!

Resumes will not be posted on bionet.jobs.offered. Such postings
should be directed to bionet.jobs.wanted.

			     IMPORTANT!!
Because of the volume of posts to this newsgroup, the moderator will
assume that the readers are familiar with the policy as stated in this
newsgroup charter and will reject without comment any messages that do
not follow the guidelines above. Appeals may be sent to
biosci-help@net.bio.net if one thinks that a posting was incorrectly
omitted. Misdirected resumes will be deleted without comment. It is
the responsibility of the posters, not the moderator, to read the
newsgroup usage instructions for bionet.jobs.wanted and
