Looking for a postdoc in protein engineering, enzymology.
I am looking for a postdoctoral fellow to take part in the study of
structure-function relationships in two related enzymes : a flavin-linked
a-hydroxyacid dehydrogenase (long chain hydroxyacid acid oxidase) and a
homologous one which is fused to a hemoprotein (flavocytochrome b2).
Present interests include : chemical mechanism of substrate
dehydrogenation, structural determinants of substrate specificity and of
the reactivity with oxygen, role of protein mobility in catalysis, inter-
and intramolecular electron transfer. Both proteins are expressed in E.
coli, a number of mutants are available, others will be made. One crystal
structure is known, the other one is being determined. Methods will include
: kinetics (steady-state and presteady-state, using substrate, substrate
analogs and flavin analogs), spectroscopy, site-directed mutagenesis,
protein chemistry, immunochemistry, etc. The program benefits from
collaborations at the national and international levels.
Our institute can be reached in less than one hour from Paris, using
convenient public transportation. It is located on a CNRS campus, in a
pleasant semi rural environment. It is equiped with molecular graphics,
microprotein sequencing, mass spectrometry, stopped-flow apparatus,
Biacore, etc.
Candidates should have a background in organic chemistry, or biochemistry
/ enzymology / molecular biology. Funding will be dependent on a
applications to the EU, EMBO, NATO or FEBS fellowships programs. There are
also bilateral agreements between the CNRS and a number of countries for
the exchange of scientists. Applicants should submit a curriculum vitae and
the names of two references.

Homeobox genes function in mammalian development. Study of Hoxa 11 and
its role in kidney and limb development. Upstream regulation and
downstream gene targets. Also study of the antisense Hoxa 11 transcript
and its function. (see Davis et al., Nature 375, 791:Small and Potter,
Genes Dev 7, 2318: Hsieh-Li et al., Dev. 121, 1375). Other possible
projects include study of homeobox genes important in brain development.
(see Scucsik et al., Dev. Biol. 191, 230), and study of a gene of
critical importance in the formation of the left-right axis of
development. (see Supp et al., Nature 389, 963). Position is located at
