POSTDOCTORAL POSITION IN CYSTIC FIBROSIS (EPITHELIAL ION
TRANSPORT)

A postdoctoral position is available immediately to study
interactions of the
cystic fibrosis transmembrane conductance regulator protein
(CFTR) with acid
-base transporters, e.g., sodium-hydrogen (NHE) and
chloride-bicarbonate (AE)
exchanger proteins. The laboratory uses
electrophysiological (Ussing chamber,
microelectrodes, patch clamp) and molecular techniques
(retroviral
transduction, RT-PCR) to investigate abnormal regulation of
transepithelial pH
and fluid transport in the disease, cystic fibrosis. Most
studies are performed on
tissues from specific transporter knockout mice (CFTR,
NHE2, NHE3) and
human cell lines. The current focus is on the mechanisms
and regulation of
bicarbonate secretion and sodium-hydrogen exchange by CFTR.

Our lab was the first to investigate the functional
consequences of CFTR gene
targeting in the mouse model of cystic fibrosis. These
studies have yielded a
number of important advances in the physiology of
epithelial ion transport,
including the identification of CFTR as the sole apical
membrane Cl- channel in
intestinal epithelia, identification of CFTR as the
principle mediator of
electrogenic bicarbonate secretion, and the discovery of
other anion
conductances with the potential to ameliorate cystic
fibrosis disease. Our
colonies of epithelial transporter knockout mice provide
unique opportunities for
reconstitution of mutant transporters into a null
background (i.e., transfection
into knockout cell lines or transgenic development of a
knockout strain) and the
study of double knockouts (e.g., interbreeding for
*CFTR:*NHE3).

Candidates for this postdoctoral (Ph.D., D.V.M., or M.D.)
position should have
interests and experience in the biological sciences
(physiology, pharmacology,
animal sciences, medicine, biology). Knowledge of
electrophysiology and/or
molecular biology techniques desirable, but not required.

Postdoctoral and Research Assistant Positions.

Applications for postdoctoral and research assistant positions are immediately available in my laboratory . Candidates should be highly motivated and have good expertise in molecular biology, biochemistry and/or cell biology techniques.
Current work underway in the laboratory focus on studying biogenic steps of G- protein coupled receptors and phosphoinositide signaling in neuronal cells. We have been particularly interested in investigating i) the molecular mechanisms underlying the function of a new neuronal signaling pathway mediated by a novel PI-phospholipase C (e.g. Ferreira, P. et al. (1994). J. Biol. Chem., 269, 3129; Ferreira, P. et al. (1993) Proc. Natl. Acad. Sci. USA, 90, 6042) and, ii) the molecular machinery involved in the intracellular processing and trafficking of homologous G-protein coupled receptors in highly polarized neuronal cells (e.g. Ferreira et al. (1998), J. Biol. Chem. submitted; Ferreira, P. et al. (1997) Proc. Natl. Acad. Sci. USA, 94, 1556; Ferreira, P. et al. (1996) Nature 383, 637; Ferreira, P. et al. (1995) J. Biol. Chem. 270, 23179). In addition, we are using our studies as a model system to study widespread homologous molecular mechanisms in other biological!
!
systems and understand the molecular pathogenesis underlying certain inherited neurological disorders in humans which ultimately lead to the degeneration of selective neurons in the CNS. The laboratory and institution are equipped with state-of-art facilities and operate in an outstanding research environment. The prospective candidate is expected to participate in campus-wide interactive programs such as seminars and ultimately, present his\her research results at international meetings and publish in top-rated scientific journals.
Candidates with an overall interest in neurobiology are invited to apply and should send their CV, summary of their past research experience and future goals, and three reference letters directly to:

Clinical Microbiologist, or Medical Technologist

Seeking someone with at least one year's experience growing
and identifying clinically important bacterial pathogens.
Your experience has preferably been in a clinical
microbiology setting (hospital, doctor's office), or, if
your background is in industry or academia, your work
should have included clinically important pathogens (as
opposed simply to bacteria of industrial interest).
However, the most important thing is that you have working
knowledge of differential media, how to "tweak" growth
conditions, and bacterial identification.

The company is located in Rockville, Maryland, and
has an active collaboration with the NIH. This is
a full-time, career position, in a growing company with
employee stock options and a pleasant work environment. We
are looking for people who take initiative, are team
players, and have flexible personalities. Salary
commensurate with experience.
