To whom it may concern,

I would like to take this opportunity to announce the availability of a Ph.D.
position in the

Department of Computer Science at the ETH in Zurich, Switzerland.

This position is associated with a new project entitled

                            Bio-Opera

and is a joint venture between the Information and Communication Systems Group
(IKS) [Prof. A. Alonso] and the Computational Biochemistry Research Group (CBRG)
[Prof. G. Gonnet, Dr. M. Hallett].

             ----------------------------------------
                        Project Description

Since 1990, the CBRG has developed a Maple-like programming environment called
Darwin which contains a robust set of tools which aid biological chemists in the
analysis of molecular data. What the system currently lacks is an organized way
to store, to retrieve, to query and, generally, to manage the large amounts of
data this analysis creates. A way of addressing this issues is to merge Darwin
with an object relational database engine. This will make Darwin a
more robust tool in terms of data management and to extend its capabilities with
the functionality offered by the database engine. This functionality includes
concurrency control for multitasking, multiuser environments, recovery, index
maintenance, constraint checking, and
optimized query processing strategies. In addition, this initial effort will
provide the infrastructure for further research in several directions:
algorithms, query languages, process support systems, and federated
bioinformatic databases.

Opera is a process management system currently being developed within the IKS
Group. It is intended as a generic kernel for the development of distributed
applications as it operates as a high level operating system in charge of
scheduling, persistence, and data consistency. Opera generalizes ideas from
federated databases, workflow management, transaction processing, and process
support, thereby providing sophisticated exception handling mechanism, data
tracking tools, automatic consistency checking and maintenance, transactional
notions for execution guarantees, and a powerful graphical environment for
developing large, complex applications.

The main goal of the project is to integrate Opera with Darwin.

             ----------------------------------------

The doctoral student would be part of a team of five working on this project.
The position begins as early as January and would lead towards a Ph.D. in
Computer Science assuming all departmental qualifications are met during the
duration of the studies. The doctoral student would receive a 100% position
within the Institute of Scientific Computing and would be working within the
CBRG group and the IKS group.

Interested parties would already possess either a Masters of Science (North
America), a Diplom (Europe), or an equivalent degree in computer science or
mathematics AND/OR molecular biology, biochemistry, or genetics. The candidate
should have an interest in databases, datastructure/algorithm theory, and a good
knowledge of genetics and molecular biology.

technology acquisitions. Identify, evaluate & assess
the compatibility & fit of a range of enabling technologies with
Pfizer's portfolio and goals. This person will focus on seeking &
establishing relationships with West Coast companies & academic
institutions. Once a technology has been identified, conduct initial
meeting(s) with the potential collaborator to gauge interest in the
technology or research as well as the fit for Pfizer. If the outcome
of that meeting is positive, conduct further due diligence, write
up findings & present to a small group internally. Facilitate
further meetings with collaborator and Pfizer scientists, managers,

Postdoctoral Position Available

We have an immediate opening for a Postdoctoral Fellow to study the role
of the thrombin receptor in cancer. In addition to its critical roles
in hemostasis and thrombosis, the thrombin receptor is likely to play a
major part in vascularization of tissue during embryogenesis and
malignant processes. The PAR1 thrombin receptor serves as the paradigm
for a rapidly developing field of protease-activated receptors which are
all activated by cleavage at a specific peptide bond by their cognate
protease. The thrombin-cleaved N-terminus of PAR1, S42FLLRN47, acts as
an intramolecular ligand that binds to the body of the receptor by a
poorly understood mechanism. Peptides corresponding to the N-terminus,
such as SFLLRN, can elicit a subset of the PAR1 responses independent of
receptor proteolysis. Three additional protease-activated receptors
have recently been identified, PAR2, PAR3, and PAR4, but their
respective roles are not well understood.

We have made the intriguing observation that high levels of PAR1
expression and activity occur in a non-small cell lung epithelial cancer
cell line. Interestingly, a recent report suggests the involvement of
the PAR1 receptor in cellular invasion associated with tumor
progression. The goals of this project include extending this
preliminary observation to see whether other cancer cell lines including
primary breast carcinoma cell lines will demonstrate a direct
correlation between the level of expression and activity of PAR
receptors and invasiveness.

Postdoctoral Position Available

Tufts University School of Medicine-NEMC

We have an immediate opening for a Postdoctoral Fellow to study the role
of the thrombin receptor in cardiovascular disease and stroke using a
small animal model system. Tissue plasminogen activator (t-PA) is a
widely used and effective agent for thrombolysis in patients suffering
from myocardial infarction. Most recently, t-PA is the first approved
treatment for selected patients with acute ischemic stroke. However, it
is clear from several large clinical trials that t-PA thrombolytic
therapy can cause severe complications such as hemorrhage, neuronal
injury, or even paradoxical activation of the hemostatic mechanism. A
concerted effort is now being made to ease the administration of t-PA,
with concomitant reduction in the incidence of debilitating
complications. Second generation t-PA drugs, such as reteplase and
TNK-tPA, are mutant forms of t-PA that lower or raise the affinity to
fibrin and hence may alter the effective plasmin concentration at other
sites such as platelet membranes or endothelial cells. The
pathophysiological basis of the most common adverse effect—namely
bleeding—is most likely due defects in platelet function. In fact, a
majority of patients receiving t-PA therapy had significant elevations
in their bleeding times regardless of whether they demonstrated
spontaneous bleeding. Treatment of platelets with low concentrations of
plasmin leads to an attenuated thrombin response, whereas treatment of
platelets with high concentrations of plasmin leads to direct platelet
activation. Our own studies indicate that these effects of plasmin are
due to truncation or direct activation of the platelet thrombin receptor
depending on the site of cleavage. An initial goal of this project is
to determine whether t-PA induced plasmin can truncate the thrombin
receptor on platelets and endothelium in animals, and if so, does
prolonged bleeding time correlate with loss of TR-dependent platelet
aggregation. These studies will be complemented by a variety of
molecular biological and biochemical approaches.
