Research Work: Elevated soil temperature constitute a major limitation to
successful crop production of tropical plants such as rice, which is an
important crop. The objective of this study  was to characterize the heat
shock response in rice leaves concentrating on the expression of heat shock
genes at the level of translation in particular.

Characterization of  the heat shock response in rice leaves under various
conditions was followed.

• Rapid decline in  cell viability and loss in the plasma membrane activity
were observed in rice leaves subjected to rapid heat shock. In contrast,
rice leaves pretreated at a mild heat shock induced tolerance to survive
better at higher temperature and the activity of the plasma membrane also
was not much affected.

• The HSP enriched fraction from rice was found to be very effective in
protecting the plasma membrane from the direct effects of heat .  The
stability of the plasma membrane bound Mg2+ - K+   ATPase in rice was found
to be proportional to the amount of post-ribosomal HSP fraction added.

• Gradual  exposure to a high temperature elicited a comparatively better
response than the   application of a sudden heat shock and heat shock
proteins were found to be synthesized even at a very high temperature of
45°C after a preincubation at a slightly lower temperature of 42.5°C which
was not possible upon  a sudden heat shock.

• The decline in the synthesis of HSPs in rice leaves was not co-ordinate.
Low molecular weight HSPs and their mRNAs were more stable and have a higher
half life than that in the high molecularweight range.

• Antibiotic studies indicated that the HSPs in rice are synthesized on
cytoplasmic ribosomes and not in the organelles and heavy metals like
Arsenite and Cadmium were found to mimic the heat shock response in rice
leaves. Etiolated rice leaves also synthesized HSPs in a temperature
dependent manner as in green leaves.

• Homologous genes and their transcripts to pea and soybean low molecular
weight HSPs were identified in rice by Southern and  Northern  analysis.

• Cross reaction of rice HSPs were observed with pea low molecular weight
antibodies on Western  analysis. Homologues to pea Class I Low molecular
weight HSP and the pea chloroplast localized low molecular  weight HSP were
identified in rice leaves.


Additional Research Interest: Regeneration  and Transformation studies in
legumes. Helped in the establishment of regeneration protocols for
regeneration of cowpea, rice and peanut from different explants and in the
successful transformation of cowpea and peanut.

Postdoctoral Positions in Molecular Biology


 NIH-funded postdoctoral positions are available immediately to study
molecular mechanisms of transcriptional regulation.  Currently there are
two on-going research programs in the lab.  One focuses on the general
transcription factor TFIIB in yeast and its role in transcriptional
activation (see MCB 16, 3652-3657, 1996; PNAS 94, 2427-2432, 1997).  The
second program addresses the questions of how the Drosophila homeodomain
protein Bicoid interacts with DNA and how Bicoid molecules communicate
with each other to facilitate cooperative DNA binding (see Development
122, 1195-1206, 1996; JBC 271, 21660-21665, 1996; Biochem. J. 338,
447-455, 1999; J. Biochem. 125, 809-817, 1999).

 The postdoctoral positions will offer competitive salary and fringe
benefits through the Children's Hospital.  Strong funding is available
for our research programs.  Our lab is physically located in the
fast-expanding Children's Hospital Research Foundation; a new research
wing was just finished and another research wing is currently under
construction.  We are a part of the University of Cincinnati College of
Medicine, where there are several research groups interested in gene
control mechanisms.

Virus Tumor Biology

Postdoctoral position available  August 1, 1999 in the Virus Tumor Biology Section, Division of Basic Sciences, National Cancer Institute. The primary research focus of the laboratory are the human retroviruses, HTLV-I and HIV. Scientific Software Engineer (ME Bar Harbor)    Position 070-99

There  is  a  regular,  full-time  Scientific  Software Engineer (SSE)
position  available  to  work  with  the  TBASE   (Transgenic/Targeted
Mutation  Database)  and  the  IMR (Induced Mutant Resources) database

A postdoctoral position is available to work on the interaction of the
sugarbeet cyst nematode (Heterodera schachtii) with Arabidopsis thaliana.
The project initially deals with the characterization of identified A.
thaliana genes that change expression around the infecting nematode.  This
gene characterization will use promoter studies, in situ hybridization,
immunolocalization, and experiments targeted to alter expression of the
gene-of-interest.  Second, this position will have an increasing genetic
component consisting of the characterization of A. thaliana mutants with
altered susceptibility to the nematode.  This genetic aspect will include
preparing and analyzing double mutants for epistasis analyses and mapping of
mutant loci.  Applicants should have strong proven records in molecular
biology, genetics, plant physiology, and techniques related to the projects
described above.  The annual salary will be $28,000 and the initial
appointment will be for one year with a renewal for additional years

Apply biomolecular NMR techniques to the diverse problems of the pharmaceutical industry including protein/peptide structure determination, implementation of state-of-the-art heteronuclear experiments; evaluation of protein-ligand interactions including binding studies and determination of Kds and application of protein dynamics to pharmaceutical problems; assist in the development of NMR program for integration of protein NMR into chemistry discovery environment including lead identification and optimization.
Two years in the pharmaceutical industry required.  Greater than three years in the pharmaceutical industry; excellent communication and teaching skills; C or FORTRAN programming; structure eludication of small molecules; familiarity with some commercial software packages including FELIX, XEASY, Discover, X-PLOR, and AMBER; and experience with maintenance of high field instruments desired.
Ph.D. with postdoctoral experience required.

This CRA position is in the CNS Therapeutic Area which includes pain, psychiatric, and neurological disorders.  The CRA is responsible for designing casebooks, working with project clinicians and teams, Contract Research Organizations (CROs), and study sites.   The CRA monitors ongoing studies and travels to the study sites to participate in initiations, review medical records, and ensure accuracy and proper adherence to protocol procedures.
Qualifications:  Bachelor?s degree required preferably in nursing or other medically related field.  Prior clinical research or CRA experience preferred.  Attention to detail, ability to work both independently and as part of a team and proficiency in Microsoft Office, PowerPoint, and Excel required.  Other desirable skills include familiarity with FDA?s rules and regulations governing clinical investigation of new drugs.
