Molecular Neurobiologists
Bai Lu, PhD

Postdoctoral positions are available to study the role of neurotrophic factors in the development and function of synapses. Neurotrophic factors are defined as secretory proteins that regulate survival and differentiation of neurons. Recent studies have revealed a novel function of neurotrophic factors in synaptic transmission and plasticity. Our lab showed that the neurotrophin BDNF facilitates hippocampal long-term potentiation (LTP), a cellular model for learning and memory (see PNAS 1994, 91: 12341, Nature 1996, 381: 706, J. Neurosci. 1995, 15: 4796; 1997, 17: 2947; 1998, 18: 6830; 1999, 19, 4972). Using a simple nerve-muscle co-culture system, we also demonstrated that long-term development of the neuromuscular synapses is regulated by the neurotrophin NT3 and other molecules (see PNAS 1995, 92: 8001, Nature 1995, 374: 262). We are now investigating the molecular mechanisms underlying the acute and long-term neurotrophic regulation. Ongoing projects include: 1) f!
unctional significance of activity-dependent BDNF expression, using transgenic/knockout mice; 2) biochemical and molecular study of activity-dependent modulation of BDNF receptor in hippocampal cultures; 3) imaging analysis of neurotrophin protein secretion and trafficking; 4) molecular study of the signaling mechanisms for acute and long-term neurotrophic regulation, using Xenopus nerve-muscle system. (see http://dir.nichd.nih.gov/LDN_Labs/LuLab/Lu_Home.htm. for detail). The candidate will be working with a team of highly interactive electrophysiologists and molecular biologists. Experience in either molecular cloning or synaptic physiology is required. A background in transgenic mice or Ca2+ imaging is highly desirable. NICHD, Building 49, Room 6A-80, MSC4480, Bethesda, MD 20892-4480, Fax: (301) 480-3526.

Pre-Clinical Research--Murine Model
Thomas Waldmann, MD

The Metabolism Branch, DCS, NCI is offering a postdoctoral research
training fellowship position for an individual who would investigate new therapeutic approaches directed toward the IL-2/IL-15 and HER2/neu receptors and their signaling pathways. The approach, focusing on the IL-2 rceptor, exploits the observation that the high affinity IL-2R is not expressed by normal cells, but is expressed by select leukemic cells as well as T-cells involved in allograft rejection and autoimmunity. A murine model of IL-2R expressing Adult-T-Cell Leukemia(ATL) was established by introducing cells from an ATL patient into SCID-NOD mice. Following serial transfer of these cells, the animals developed grossly enlarged spleens and leukemia of ATL cells that retained the phenotype and HTLV-I integration pattern of the patient''s ATL leukemia. The candidate would be involved in the development of novel therapeutic agents and in their evaluation in the mouse ATL model as well as in a model of HER2/neu expressing breast cancer. Agents showing promise in these muri!
ne models may move forward into clinical trials. Humanized anti-receptor antibodies armed with alpha and beta emitting radionuclides or with toxins, as well as JAK-3 kinase-directed inhibitors, are some of the agents under investigation. Applicants should have completed an M.D.
or Ph.D. and be highly motivated individuals interested in joining a
progressive Clinical Research Program. The Metabolism Branch, DCS, NCI, Building 10, Room 4N115, 10 Center Dr MSC 1374, Bethesda, MD 20892-1374.

Please tell your students and colleagues about the following
postdoctoral position:

STRUCTURAL ANALYSIS OF SINORHIZOBIUM LIPOPOLYSACCHARIDES AND EVALUATION
OF THE STRUCTURAL FEATURES REQUIRED FOR SYMBIOTIC INFECTION
The University of Georgia Complex Carbohydrate Research Center (CCRC)
has an immediate opening for a POSTDOCTORAL RESEARCH ASSOCIATE to
conduct research on the cell surface lipopolysaccharides (LPS) from
Sinorhizobium, a major symbiont of legumes. The LPS are dominant
surface antigens of these bacteria and are believed to play a key role
in the process of nitrogen fixation. The long term goal of this
research is to determine which LPS structural features are essential for
proper symbiotic infection, and to determine if LPS structural
alterations, which occur during differentiation, help determine host
range/specificity. Structural analysis of these glycoconjugates
involves the use of mass spectrometry (electrospray ionization,
MALDI-TOF, FAB) and standard 2D NMR analyses of the intact molecules and
their subunits, in addition to specific chemical methods of
polysaccharide degradation and derivatization in combination with GLC-MS
and HPLC techniques. Candidates should have some previous experience
with at least one of these techniques, in addition to a strong interest
in the structure/function of glycoconjugates. Experience with all of
these techniques is not a prerequisite. Support is available for
approximately a 3 year period, and the salary will be based on previous
training and experience. Please send a letter of application, names and
addresses of three references, and a current Curriculum vitae to Dr.
Lennart Scott Forsberg, Complex Carbohydrate Research Center, The
University of Georgia, 220 Riverbend Road, Athens, Georgia, 30602-4712,
USA. Position open until filled. An EEO/AA institution.

Thank you

Scott Forsberg

Clinical Research

Two postdoctoral positions are available immediately in the Center for
Computational Biology at Montana State University, Bozeman, in the
laboratories
of John Miller and Gwen Jacobs. Both positions are to study information
processing in the cercal sensory system of the cricket. Applicants for
both
positions should have experience in electrophysiology.

The focus of the project in Miller?s lab is to understand encoding of
dynamic
sensory stimuli by ensembles of nerve cells. The primary experimental
approach
will be multi-unit extracellular recording. Candidates should have
background in
the application of information theory to analysis of neural systems. The
focus
of the project in Dr. Jacobs? lab is to understand the physiological
mechanisms
underlying neural encoding in this system. Experimental approaches will
include
intracellular and optical recording, and advanced morphometric analysis
of
identified nerve cells. For both projects, analysis of the data will
involve the
development of compartmental and analytical models of identified nerve
cells and
networks.

Advanced computational, microscopy and physiological recording
facilities are
available within the Center, including a 32-processor SGI Origin2000
computer
and a Leica TSP confocal microscope. More information about the Center
for
Computational Biology can be found at: www.nervana.montana.edu.

Molecular Biologist
Doug Brenneman, PhD

The SDMP has an immediate opportunity for a molecular biologist with a
Ph.D. The research program has focus on the role of neuropeptides in
neurotrophism and embryonic growth regulation. A new class of
neuroprotective and growth-regulating molecules has been discovered that protect central nervous system neurons from clinically relevant
neurotoxins. An experienced molecular biologist is needed to work with a multidisciplinary research team to clone and characterize substances related to the activity dependent neurotrophic factor family of proteins and to discover neuropeptide receptors that mediate growth factor secretion. Knowledge of expression cloning and PCR is essential. This is a two-year IRTA fellowship that may be extended up to five years. Starting salary is commensurate with experience. Section on Developmental and Molecular Pharmacology, MICHD, Building 49, Room 5A38, Bethesda, MD 20892.

MOlecular Epidemiology and Immunology
Margaret C. Bash, MD

The major outer membrane porin proteins of the pathogenic Neisseria species are being investigated. The role of PorA in stimulating a protective immune response to Neisseria meningitidis is well supported. It remains unclear whether or not the gonococcal porins PIA or PIB proteins can stimulate protective immunity. The molecular epidemiology of gonococcal por variable regions (VR) among clinical isolates will be determined utilizing a VR genotyping system. Systemic and mucosal immune responses to corresponding porin protein antigenic epitopes will be measured. These assays will require the development of hybrid porins and or deletion mutants. Position available immediately. Candidates must be U.S. citizens or permanent residents. Applicants must have less than five years of postdoctoral experience. Funding: FDA Office of Women''s Health grant through the ORISE program. Laboratory of Bacterial Polysaccharides, Center for Biologics Evaluation and Research, Building 29, (NI!

Looking for a postdoc in protein engineering, enzymology.
I am looking for a postdoctoral fellow to take part in the study of
structure-function relationships in two related enzymes : a flavin-linked
a-hydroxyacid dehydrogenase (long chain hydroxyacid acid oxidase) and a
homologous one which is fused to a hemoprotein (flavocytochrome b2).
Present interests include : chemical mechanism of substrate
dehydrogenation, structural determinants of substrate specificity and of
the reactivity with oxygen, role of protein mobility in catalysis, inter-
and intramolecular electron transfer. Both proteins are expressed in E.
coli, a number of mutants are available, others will be made. One crystal
structure is known, the other one is undergoing refinement. Methods will
include : kinetics (steady-state and presteady-state, using substrate,
substrate analogs and flavin analogs), spectroscopy, site-directed
mutagenesis, protein chemistry, immunochemistry, etc. The program benefits
from collaborations at the national and international levels.
Our institute can be reached in less than one hour from Paris,
using convenient public transportation. It is located on a CNRS campus, in
a pleasant semi rural environment. It is equiped with molecular graphics,
microprotein sequencing, mass spectrometry, stopped-flow apparatus,
Biacore, etc.
Candidates should have a background in organic chemistry, or
biochemistry / enzymology / molecular biology. Funding will be dependent on
aapplications to the EU, EMBO, NATO or FEBS fellowships programs. There are
also bilateral agreements between the CNRS and a number of european
countries for the exchange of scientists. Applicants should submit a
curriculum vitae and the names of two references.

Molecular Population Geneticist

National Exposure Research Laboratory - Cincinnati
U.S. Environmental Protection Agency

A postgraduate research fellowship is available to work in the Molecular
Ecology Research Branch of the National Exposure Research Laboratory
(NERL), located in Cincinnati, Ohio. With guidance from a research
mentor, the research fellow will develop codominant molecular markers in
order to (1) characterize genetic diversity of freshwater aquatic
species residing in diverse environments and (2) study the relationship
between genetic diversity and indicators of anthropogenic stress. The
researcher will be allowed a high degree of autonomy to develop and
pursue an original plan of research that addresses these broad
objectives. Examples of relevant topics include:

- Development of single-nucleotide polymorphism or microsatellite DNA
markers, and development of screening methodologies based on
high-throughput capillary electrophoresis or DNA chip technology.
- Screening of natural populations with panels of codominant DNA markers
to estimate population genetic parameters.
- Isolation of DNA markers linked to genes involved in adaptation to
contaminant-exposed environments.
- Experimental analysis of the importance of genetic diversity to the
survival of populations of aquatic organisms following exposure to novel
stressors.
- Application of analytical methods that effectively integrate
information from diverse classes of molecular markers (e.g., cytonuclear
disequilibrium analysis) to provide novel insights into population
genetic processes.

The Molecular Ecology Research Branch of NERL utilizes state-of-the-art
equipment and laboratory facilities to develop and assess molecular
indicators of the health of the nation's aquatic ecosystems. It is
expected that the research fellow will acquire unique educational
benefits by developing and executing this plan of research at
NERL-Cincinnati and through interaction with US EPA scientists using
diverse ecological monitoring methodologies.

Qualifications

The successful candidate will be highly skilled in the use of modern
molecular biology/genetics methodologies and in their application to
ecological and environmental issues. He/she will be able to
conceptualize and execute an original plan of research, establish
research priorities and deadlines, judge the completeness and accuracy
of research results and present his/her research findings to diverse
audiences. An M.S. degree in related subject matter is expected; PhD.
preferred.

The appointment is for one year and may be renewed subject to
availability of funds.

The Postgraduate Research Participation Program for NERL is administered
by the Oak Ridge Intitute for Science and Education (ORISE). Potential
applicants are strongly encouraged to contact Dr. Mark Bagley
(bagley.mark@epa.gov, 513-569-7455) for details about this research
opportunity. For additional information on the ORISE program and for
