Post Doctoral/Research Associate Position
We have used the glycoprotein hormones as a model system to reveal the
role played by unique forms of terminal protein glycosylation that
convey information for cellular recognition. This has led us to define
a remarkable system in which the same unique sulfated carbohydrate
structure on different glycoproteins plays critical biologic roles in
reproduction, development, and the immune response. The sulfated
sugars are recognized by a complex receptor with multiple carbohydrate
specificities. When expressed in liver the receptor regulates the
half-life of hormones in the blood bearing these structures. When
expressed on the surface of other cells the same receptor mediates
cellular recognition through membrane glycoproteins bearing these
sulfated sugars. Expression of the enzymes that add the sulfated
sugars to glycoproteins as well as the receptor that recognizes them
are hormonally and developmentally regulated. We are utilizing
biochemical analysis, mutagenesis, an!

d genetic manipulation of the cloned transferases and receptor to
examine the biology of this unique form of glycosylation in vivo and
in vitro. I have openings at number of different levels for
individuals interested in advancing this story.

Postdoctoral Fellowship: Bacterial Pathogenesis/Host Response
Our laboratory is seeking a highly motivated and talented individual
to join our basic research program on group B streptococcal (GBS)
pathogenesis. GBS are the leading cause of pneumonia, sepsis and
meningitis in human newborn infants. The project, funded through a
two-year grant from the United Cerebral Palsy Research Foundation,
willl explore the early steps in the pathogenesis of GBS meningitis.
Our model system examines the interaction of GBS with human brain
microvascular endothelial cells (BMEC), the single cell layer
comprising the blood-brain barrier. [see Infection & Immunity 1997;
65(12): 5074-81]. Using various mutagenesis and transformation
strategies, we are in the process of identifying specific virulence
determinants which allow GBS to invade, transcytose or injure this
cellular barrier. We are also particularly interested at the moment in
beginning to characterize the acute (predominantly neutrophilic)
inflammatory response which the bacteria, and specific bacterial
factors (e.g. hemolysin), elicit at the site of infection. These
studies will examine cytokine release (IL-8, TNFa, IL-1a) upregulation
of neutrophil adhesins in the BMEC exposed to GBS.

The position is available beginning July 1st. A candidate with a
background in pathogen/host cell interactions, Gram-positive molecular
genetics, or cytokine responses to microbial pathogens is preferred.

Postdoctoral Researcher-Yeast Cell Biology, Syracuse University
I am seeking a postdoctoral researcher to aid in our investigations of
the yeast mating pathway. Specifically, I would like to recruit a
postdoctoral researcher to study a pheromone-induced cell wall protein
encoded by the FIG2 gene. Projects include genetic, cell biological,
biochemical and genomic approaches to determining the function of the
Fig2 protein in yeast mating. Because the fungal cell wall is a choice
point of attack in the design of antifungals by the pharmaceutical
industry, it is likely that experience in this area will provide the
interested researcher with an excellent background suitable for either
academic or industrial career paths in the biomedical sciences.

Post doctoral Associate
LOCATION: Department of Pharmacology, Medical School, Academic Health
Center, University of Minnesota.

REQUIREMENTS: Ph.D. in one of the biological sciences. Experience in
mammalian cell culture, cell-cycle regulation, enzymology, protein
purification, and molecular biology, particularly desirable.

PROJECT: Identify the "molecular determinant(s) of cellular
sensitivity to flavopiridol, an anti-cell signaling anticancer agent."
Primary determinants of cellular sensitivity to flavopiridol are
likely to be cellular levels of cyclin-dependent kinase activities.
Alternatively, cellular levels of relevant drug-detoxifying enzymes,
drug transport pumps and/or nontarget flavopiridol-binding proteins
may be operative. Initial studies will be of a cultured subline of the
human breast adenocarcinoma MCF-7 cell line that is stably 24-fold
less sensitive to flavopiridol than is the parent line.

SALARY: Negotiable.
APPLICATION DEADLINE: Open until filled.
BEGINNING DATE: August 1, 1999 or thereafter.

Postdoctoral Positions
Several postdoctoral positions are available at the Albert Einstein
College of Medicine. The areas of interest include:
1. Inhibitor Design. The design and analysis of tight binding
transition

state inhibitors for the hydrolases and phosphorylases involved in
nucleoside and nucleotide metabolism are being pursued with Vern
Schramm

Albert Einstein) and Charles Grubmeyer (Temple University). These
projects are directed towards the development of therapeutic agents
for a

number of medically relevant areas, including malaria, sleeping
sickness,

graft rejection and tumor suppression.
2. Synthetic Blood Substitutes. The design of synthetic blood
substitutes is being pursued with Chien Ho (Carnegie Mellon
University).

This program combines high resolution crystallography, NMR
spectroscopy

and site directed mutagenesis for the development of mutant
hemoglobins

that demonstrate low oxygen affinity, high cooperativity and stable
tetramer formation in a cell free environment.
3. Immune Response. In collaboration with Stan Nathenson (Albert
Einstein) we are investigating the structural mechanisms responsible
for

regulation of the immune response. In particular, there is
considerable

focus on understanding the biological role of the numerous
co-stimulatory

factors involved in T-cell activation; the mechanisms of antigen
presentation by the immune system; and the mechanisms by which viral
pathogens escape surveillance by the immune system. In addition to
crystallographic analysis this program provides ample opportunity to
gain

experience in eukaryotic protein expression systems and protein
chemistry.

4. Structural Genomics. In collaboration with Rockefeller University
and

the Biology Department at Brookhaven National Laboratory we have
established a program for high throughput protein expression,
purification, crystallization and structure solution. The progress of
this program can be viewed on our website at
All of these positions provide the opportunity to acquire skills in
molecular biology, protein expression, protein chemistry, spectroscopy
and

cell biology. The Albert Einstein College of Medicine and the greater
New

York area provides an extraordinarily strong environment for
structural

biology. The College administers two beamlines, X9A and X9B, at
Brookhaven National Laboratory with dedicated MAD capabilities. The
College also has outstanding facilites for NMR, time-resolved
fluorescence, IR and resonance Raman spectroscopies. The New York
Structural Biology Center located in NY City will house the largest
NMR

facility in the country, including six 800 MHz and five 600 MHz
spectrometers.
