Postdoctoral Positions in Disease-Associated Defects in RNA Metabolism

Postdoctoral positions are available to study the mechanism by which
nonsense codons elicit mRNA decay in mammalian cells. Nonsense codons,
caused by either frameshift or nonsense mutations, are responsible for
an estimated one-third of inherited genetic diseases. If the resulting
premature termination of translation can be abrogated even slightly so
as to increase the amount of full-length protein synthesized, then the
abnormal phenotypes of many diseases can be abrogated. Therefore, an
understanding of nonsense-mediated decay (NMD) is critical to the
development of clinically useful therapies.

Currently, we are interested in understanding how nuclear pre-mRNA
processing and other nuclear events influence mRNP structure in ways
that impact cytoplasmic mRNA translation and, in so doing, NMD. Studies
will focus on various splicing and translational factors that we have
already found influence, respectively, mRNP structure and NMD in order
to establish how these factors interact and function (see, e.g., Sun
and Maquat, 2000, RNA 6:1-6; Le Hir et al., 2000, Genes & Dev. 14:1098-
1108; Sun, Moriarty, and Maquat, 2000, EMBO J, 19:4734-4744).
Interestingly, some of the translational factors derive from
alternatively spliced pre-mRNAs so that different isoforms may have
different interactive capabilities and, therefore, different roles in
NMD. Having demonstrated that NMD involves a PI 3-kinase-related kinase
signaling pathway, we are currently pursuing studies of the putative
kinase and its modifiers. We are also interested in understanding why
certain nonsense-containing mRNAs are immune to NMD decay as well as
the mechanisms that specify two distinct cellular sites for NMD.

Significant projects are available in molecular biology, biochemistry
and cell biology. Successful candidates will join a well-equipped group
of interactive lab members with diverse backgrounds and expertise in
newly remodeled labs. The University of Rochester is unique for its
sizeable community of RNA researchers and its interest in clinical
approaches to modify disease-associated gene expression by modifying
RNA metabolism.

Postdoctoral job position posted on the Post-doctoral website:

     http://www.Post-docs.com

=============================================

Two Research Associate positions are available to study cellular and
molecular mechanisms underlying salt sensitive hypertension. In
particular, we study the molecular events that participate in tissue-
specific regulation of angiotensin receptor subtypes in a variety of
models of hypertension and diet manipulations. Furthermore, we have
recently developed a novel animal model of salt sensitive hypertension
that is sensory nerve-dependent. This model provides a novel
experimental paradigm for exploring underlying molecular mechanisms
linked with salt sensitivity and sensory function. Interested
individuals should send their curriculum vitae and names of three
references to: Dr. Donna H. Wang, Professor of Medicine and Director of
Investigative Medicine, Michigan State University, Department of
Medicine, B316 Clinical Center, 138 Service Road, East Lansing,
Michigan 48824. Fax: 517-432-1326.

protein crystallography purposes. Applicants should have experience in
molecular biology and/or protein expression/purification. The lab
is well equipped with the tools for bacterial, yeast, and mammalian cell
culture protein expression. In addition, we have BIOCAD Sprint, FPLC,
and AKTA-FPLC purification systems.
This position offers also a unique opportunity to learn new state-of-
the-art structural biology techniques such as crystallization,
data-collection and more if interested.

In addition, post-doctoral opportunities for biochemists who have
an interest in structural biology are available as well. The successful
applicants will join a strong and internationally recognized Structural
Biology Center (see van den Akker et al. Nature 406, 101-104, 2000) that
is aiming to be at the forefront in this genomics age of research.
The aim of the project in the lab is to elucidate the molecular mechanism
of several guanylyl cyclase coupled receptors which are involved in a
variety of physiological processes such as blood pressure regulation and
photo adaptation in cones and rods.
The successful candidate should have experience with handling proteins
in particular their over-expression and purification. The applicant
should also have an interest in structural biology and a desire to
learn protein crystallography but experience is not required.
Our laboratories are located in state-of-the-art facilities in the Lerner
Research Institute and are fully equipped for E. coli, yeast, and
mammalian cell protein expression, protein purification, X-ray diffraction
and computation. For more information about our research please visit our

US-IL, Postdoctoral position for functional studies of MLL fusion
proteins in leukemia

Postdoctoral positions are available for functional studies of MLL
fusion proteins in leukemia.

NIH funded research projects focus on elucidating the mechanisms
whereby MLL-CBP and MLL-AF9 cause leukemia. We will use two model
systems to study the role of normal MLL and MLL fusion proteins on the
chromatin structure of HOX target genes, to identify additional target
genes, critical protein functions and protein-protein interactions. We
have established a retroviral mouse model of MLL-CBP leukemia and are
in the process of developing an in vitro inducible tissue culture cell
model that we will use to identify downstream target genes (Chip and
microarray technologies) and critical protein functions.

Relevant papers: PNAS 94, 8732, 1997; Blood 90, 535, 1997; PNAS 91,
10610, 1994; Blood 87, 1912, 1996; Blood 92, 3793, 1998; PNAS 96,
6428, 1999.

Experience in any or all of the following areas is desirable: protein
biochemistry, chromatin/nucleosomal remodeling, molecular biology, RNA
analysis, tissue culture.

Candidates must have a Ph.D. or an M.D. degree.
Salary commensurate with experience.
Interested individuals should send a c.v. and the names of three
references via email or mail.

                   Royal Botanic Garden Edinburgh

The Royal Botanic Garden Edinburgh (RBGE) has a vacancy for a Higher
Scientific Officer (pensionable Salary 17,200 pounds sterling) for a
three year BBSRC funded project exploring the capturing and relating
of character concept definitions in plant taxonomy. This is a joint
project between the RBGE and the Database and Object Systems Group of
Napier University. The successful candidate will join an already
successful team of bio-informatics specialists working to improve the
rigour and quality of taxonomic data storage and dissemination.

The work will involve close collaboration with the Napier team to
formulate and refine a generalised character space model that can be
used to capture any character concept. The successful candidate will
be expected to help produce and test this model and use it to create a
foundation set of character concept definitions. We intend to cover a
wide range of materials and the candidate will work with data ranging
from the most up to date molecular results through to examination of
traditional morphological characters such as those found in standard
floras and monographs.

Candidates must have an M.Sc. or Ph.D. in taxonomy and relevant
experience in identification and the use of character concepts.

More details of the current project proposal and the earlier
Prometheus project can be found at www.prometheusdb.org

Applications, including a full C.V. and details of two referees
should be made in writing to Dr. Martin Pullan, The Royal Botanic
Garden Edinburgh, 20A Inverleith Row, Edinburgh EH3 5LR, or via email
to m.pullan@rbge.org.uk before 5th January 2001.
