How to reply to a message on the group:

Dear Mary:

This posting is not about a specific technique, but about something that I've
been meaning to bring to the attention of the editors of your journal for some
time. Many of the printed articles are interrupted by advertisements and since
not all the pages of BioTechniques have the page numbers on them, photocopied
articles are difficult to reference without looking them up in the original
volume. This can be very frustrating when a number of articles are to be
referenced at one time, forcing one to go to the library and look all of them
up again to be sure of the correct page numbers. It would be a vast improvement
if the numbers were printed on ALL pages, or the page numbers for the reference
printed on the first page of the article itself as many other journals do.

I am soon to embark on immunohistochemical techniques to see specific 
proteins in tissue samples. Having never used these techniques before, I 
was wondering if people would share their preferences with regards to 
using cryo-sectioned tissues, cryo-sectioned tissues--and then fixed, and 
wax embedding tissues--usually using paraformaldehyde prior to embedding.

Which do people prefer? And from personal experience, which have led to 
more successful preparation in the end? 

I will be analyzing the aorta, heart, liver and spleen
