Our laboratory studies the class of cell-surface protease-activated
receptors (PARs). PARs are members of the receptor super-family that
signals through intracellular switches called G proteins. The
first-identified protease-activated receptor, PAR1, is present on
platelets, blood vessels and invasive cancer cells. PAR1 is activated
when thrombin cleaves the peptide backbone at a specific site located
within the receptor N-terminal extracellular domain. PAR2 and PAR4 are
activated by other serine proteases present in blood and tumors.
Proteolytic cleavage of the PARs exposes a new N-terminus that activates
the receptor in an unusual intramolecular mode. There is much interest
in developing reagents that can specifically inhibit the PARs and
prevent heart attacks, stroke, inflammation, pain, asthma, and
metastasis of cancer cells (Cancer Research (2001) 5933-5940).

With this goal, we recently discovered (PNAS (2002), 643-648) a new
class of cell-penetrating, membrane-tethered peptides—termed
pepducins—that are based on the intracellular loops of the PARs.
Pepducins rapidly pass through the plasma membrane of intact cells and
cause full activation or inhibition of G protein-dependent signaling.
Most remarkably, pepducins are selective for their cognate receptor and
require receptor for activity. We have shown that these novel
cell-penetrating peptides are potent inhibitors of receptor-G protein
signaling under in vivo conditions in live mice. The post-doctoral
fellow will study the mechanism of action of the pepducins using a
number of approaches including peptide chemistry, biochemistry, and
molecular biology of mammalian cells expressing the PARs. Fluorescence
resonance energy transfer (FRET) will detect interactions between PARs,
pepducins and intracellular effectors.

Our laboratory is located within the Molecular Cardiology Research
Institute, a state-of-the-art facility supported by several core units
including peptide synthesis, DNA sequencing, plasmid preparation, small
animal surgical and physiology lab, flow cytometry, and transgenic mouse
facility. Qualifications for this position are a Ph.D. degree;
experience in mammalian cell transfection and expression is preferred.

US Census 2000 data is now available from GeoLytics

The U.S. Census Bureau has recently released the first 
two major sets of data from the 2000 Census: 
Redistricting and the "Short Form" (SF1). 

GeoLytics offers these products in two easy-to-use formats 
either at the Block level or at the Block Group level 
and larger geographies.  The data has been compressed 
so that it generally fits on 1 disk and it comes with 
built-in mapping capabilities, based upon the new 
Census 2000 boundaries.  With a few quick keystrokes
