Jilko--

I am interested in bioinformatics.  I recently participated in
2 courses/workshops at the University of California-San
Diego in the subject.  I am also currently studying C++ and
Visual C++ for the purpose.

There are easier programming languages to learn, but I like
a challenge.

Is this an appropriate newsgroup for a discussion like this?
Does anyone have another suggestion?

The Law will soon bow to Genetics

    As the saying goes "Individuals donot evolve* but population do". We are
imprisoned in the Genetic materials that our parents gave unto us through
the unity of their  haploid gametes to produce the diploid us. At present,
the Human Chromosomal Mapping is on
going by the developed countries.. Subsequently as I believe the phenotypic
bearing of each gene will soon be revealed though regulatory (the extent of
its power to promote or its constitutive flow , etc.) or structural. Genes
that are environmentally prone for their
activities will soon shed light to the badness and goodness of an
individual. Phenotypically bad because of  his/her bad/poor or the like type
of milieu.. Phenotypically good because of his "good"/rich environment where
bad characters could be masked as they could be bought by cosmetic genetics
( behavioral or physical). But
who these  good/learned  individuals say that they have the power to judge
the poor in their own rich living room...the influence to their genes which
mold their character are not the same as that of the poor/bad... Genetics
will soon sum up the totality of an
individual and will reprieve individuals from the penalty that law has
unfairly given them.

*Our lifespan is too short to accommodate natural selection.

several papers seem to show a pathogenic effect  of drugs(namely cannabis)
on the animal, or on the human genome .several others don't.The advise of
the experts on drugs and the maining about this problem from the geneticians

My deceased wife worked for a Genetic Center  on Long Island.  She might be
able to help you for she is an expert in the field of translocated chromosomes.
 You would, however, have to know the exact breakpoints on chromosomes 4 & 18.

Hello, my name is Alex and at the moment I am in my second year of
university at Reading, UK.  Myself and 4 others are currently trying to

Depends what machine and newsreader. EG this is a Mac and running the old
Newsreader. I have an Acorn Risc also running newsbase/ttfn with Drag and

The male sex determining gene is located on the short arm of the Y chromosome. 
However there are XX males where the male sex determining gene appears to have
been been relocated to the X, and XY females where the males sex determining
factor has been removed or inactivated.  And ther are other exceptions.

suppose that u discover two interesting rare cytological abnormalities in
the karyotype of a human male. There is an extra piece(sattelite) on one of
the chromosomes of pair 4, and there is an abnormal pattern of staining on
one of the chromosomes of pair 7.
  assuming that all the gametes of this male is equally viable, what
proportion of his children will have the same karyotype as he has?

Functional genomics/molecular genetics
of Phytophthora infestans

Postdoctoral associates are sought to study Phytophthora infestans, the
oomycete fungus that causes the late blight diseases of potato and tomato.
P. infestans is one of the world's most important plant pathogens, with a
direct economic impact exceeding $5 billion per year.

The successful applicants will join projects aimed at identifying and
characterizing genes important in the growth and development of P.
infestans, with an emphasis on sexual and asexual sporulation.  The
projects involve constructing cDNA libraries; identifying genes of interest
from an EST catalogue and an expression profiling database (for example,
microarrays); and characterizing the regulation and function of genes by
manipulating their expression in P. infestans transformants and other
strategies.  Funding for these positions are available for up to 3 years.

The laboratory is located at the Riverside campus of the University of
California, which is situated in a dynamic region of southern California.

Our daughter was born 10 months ago with a deletion of chromosome 22q with a
breakpoint at 13.2 does anybody have any information about this problem, The
doctors are helpfull but not very informative.  she at the moment is a
healthy young girl but we as parents worry about her future.

As of 10 December 1995, all BIOSCI/bionet full newsgroups are
accessible through the World Wide Web (WWW) at URL http://www.bio.net.
One can read and reply publicly or privately to both recent postings
and archived messages through one's Web browser if it is configured
properly to send e-mail.  Each newsgroup is equipped with its own WAIS
index.  The main BIOSCI home page also has access to the BIO-JOURNALS
Table of Contents database WAIS index and the BIOSCI user address
database described in another item further below.


2) What to do about "spams," i.e., junk mail, ads, etc.
-------------------------------------------------------
BIOSCI is a set of parallel USENET newsgroups (the "bionet" groups),
mailing lists, and a hypermail archive at URL http://www.bio.net/.
The same postings are distributed on all media (except for a small
number of mailing-list-only groups at net.bio.net).  Unfortunately it
is becoming a despicable practice on the Internet (by a few people out
to make a fast buck) to do automated mass postings to thousands of
newsgroups and mailing lists.  These attempts to grab free advertising
are refered to as "spams" in the usual, somewhat boneheaded, net
terminology.  USENET is more susceptible to this practice, and many
spams originate on the USENET groups and then are passed on to the
mailing lists.  However, spammers also get lists of mailing addresses
and hit these too, so neither medium is immune.

What should you do personally if you get junk mail?
---------------------------------------------------
Just delete it and move on without reading it further.  Filing a
protest is becoming increasingly useless because spammers are often
disguising the addresses where the messages are sent from.  Unless you
really understand Internet mail systems, your attempt at protest by
sending replies to the message will often end up being sent to the
address of an innocent person that the spammer is victimizing.

What can BIOSCI/bionet do to protect its newsgroups?
----------------------------------------------------
The only solution currently available is to moderate the newsgroup.
If this newsgroup is already moderated, then you are in good shape.
Moderation protects the USENET distribution from about 95% of the
spams that are being sent to date and protects the mailing lists
completely.  Moderation means, however, that someone has to take the
time to review each message before it goes out.  We have set up
software here that simply allows the moderator to forward to an
address at net.bio.net messages that (s)he wishes to have distributed.
This takes no more time than that needed to read the message and pass
it on, say about 1 min. per message.

Most newsgroups currently have a discussion leader who is responsible
for their newsgroup.  The discussions leaders and their e-mail
addresses are listed in the BIOSCI Information Sheet which is
available on the Web at http://www.bio.net/.  If a newsgroup is being
hit with too many junk postings, please contact the discussion leader
for that group and see if there is interest in moderating the group.
Please do not assume that by simply posting a complaint to the
newsgroup itself, anyone on the BIOSCI staff will act on your
complaint.  With close to 100 newsgroups to run, the BIOSCI staff has
to rely on the discussion leaders of each newsgroup to report problems
directly to us at biosci-help@net.bio.net.

We will moderate any of our newsgroups if the discussion leader tells
us that the readership of the group wishes to do so and if a moderator
is willing to do the work.  For most BIOSCI/bionet groups, this
entails only a few minutes of work each day.

Moderating a newsgroup will resolve probably 95% of the junk postings
on the USENET distribution.  Unfortunately there are easy ways for
determined spammers to override the moderation mechanism on USENET,
but we can protect our e-mail subscribers from unwanted postings if
the newsgroup is moderated.  You can also access our newsgroups over
the WWW at URL http://www.bio.net.  While this Web interface will not
stop spammers from trying to post to the groups, this will give you
yet another way, besides using USENET news, to keep the junk out of
your personal mail files.  For those of you with local USENET news
systems, the Web interface will also give you faster access to new
newsgroups and recent postings.


3) Examples of subscribing and unsubscribing to the mailing lists.

PLEASE NOTE: The BIOSCI management does NOT act on
subscription/unsubscription requests that are posted improperly to the
newsgroups and mailing lists.  People who do this only bother everyone
on the lists to no avail.  Please be sure to follow the proper
procedures below.

http://www.bio.net.  Below we give an example utilizing the
METHODS-AND-REAGENTS list at both of our two BIOSCI sites:

B) Mail all commands in the body of a mail message addressed to
   biosci-server@net.bio.net.  Do NOT send commands to the newsgroup
   posting addresses!  Leave the Subject: line blank, any text on it
   will be ignored.

C) In the body of your message put one or more of the following
   commands with an "end" command on the last line, e.g.,

   subscribe methods
   unsubscribe methods
   end

   Do NOT put your e-mail address or other text on these lines.  The
   server only allows you to cancel your subscription if the address
   on your mail header matches the address on our mailing list.
   Please ask for help at biosci-help@net.bio.net if your address has
   changed, e.g., if you know you are on the list but the server tells
   you that you are not a member.

It depends on  the species!

In humans,  XX is a normal female and XY is a normal male, so the "Y" is
the "male" chromosome.  (That is, of course, a simplification!)

For other species,  the males have two sex chromosomes of the same type
and it is the female that has one different (birds, for example).

The H63D and C282Y    HFE mutations are the two know mutations causing
iron overload in humans
(due to a dysfunction in iron regulations - specifically, a failure to
down regulate the uptake of iron even
when excess iron is already present.)

I am a member of a web support group for patients with this disorder.
It seems apparent from many
postings on our list that the clinical presentation of those with one or
two H63D alleles  is very different
than than of the far more common homozygous state for the C282Y
mutation.       The major difference
appears to be that iron accumulates and causes damage in tissues other
than the liver even *before*
excess levels have begun to accumulate in the liver.   (with the C282Y
alleles,    iron does not begin to
accumulate in other tissues until a large excess has accumulated in the
liver.)

I feel that this difference has been missed clinically for the following
reasons:

Until very recently (i.e.  the location of the gene involved) it was not
possible to distinguish between the
two mutations.    Because the H63D allele is now known to have a much
lower frequency than the C282
mutation,    the clinical experience of iron overload and the
development of diagnostic standards and
therapy was based almost entirely on observation of patients homozygous
for the C282Y mutation.
(Any differences in lab values,  clinical presentation, etc. in the H63D
patients would have almost
certainly be attributed to supposed co-existing unidentified medical
problems (because of the rarity of
such differences).   Since there would also, of course, be a number of
other coexisting medical
conditions in many patients,    any consistent pattern of differences in
H63D patients would be unlikely to
be noted due to the confounding effect of other conditions.

In addition,  if,   as now seems to be the case,    symptoms and
problems occur in H63D patients at
much lower levels of iron accumulation (and possibly even with no
accumulation at all) H63D patients
would be much less likely to be even diagnosed as having a disorder of
iron regulation and thereby not
even be considered in the same group as C282Y patients.    (Unless serum
ferritin, for example, is very
elevated,  iron dysregulation is unlikely to be even considered as a
possible diagnosis.    Yet some H63D
patients experience major problems even at low levels of ferritin.)


I think that the probable structural differences between the effects of
the H63D and the C282Y
mutations give good reasons for suspecting that major differences
between the effects of the two
mutations would be likely:      H63D replaces a histidine residue
(basic) with an aspartate residue
(acidic),  the shape of the replacement is very different (a side chain
vs a side ring),  the replacement has
only 2 pKa's whereas the original has 3,  and the lost histidine tends
to have strong enzymatic activity.
But the C282Y mutation mainly has the effect of the loss of a disulfide
bond - leading to loss of stability,
with the possibility of partial functionality remaining.

I have only part of a biochemistry course, so I might well be wrong
about that.  Does this seem
reasonable?   I would welcome any comments!

Does anyone on this list know of significant differences between the
regulatory effects of the H63D and
the C282Y   HFE mutations?


(Please e-mail me direct as I will not have time to go through all the

list seems to be highly appropriate I am sending it to groups that are
as closely related as I can find.)

Early resolution of this issue might make a profound difference in the
health of many H63Ds for who
current diagnosis and treatment seems to be clearly inadequate for
whatever reason.
