Next to routine TB diagnosis (microscopy, BACTEC and other conventional 
culture/biochemical typing procedures, PCR, NASBA) we are currently 
evaluating a number of RFLP protocols for epidemiological purposes. 

Here are some of the corresponding publications and principles:

(van Embden et al., J. Clin. Microbiol. 31, 406-409, 1993): PvuII cleavage of 
genomic DNA and subsequent hybridization with an IS 6110 specific probe. 

(Ross and Dywer, J. Clin. Microbiol. 31, 329-334, 1993): Amplification of 
DNA between clusters of IS6110 on the genome.

(Friedman et al., J. Clin. Microbiol. 33, 1383-1384, 1995): Amplification of 
DNA between polymorphic GC-rich repetitive sequences and IS6110 elements.

However, this advanced protocol is our favourite: 
(W. Haas et al., Mixed-linker polymerase chain reaction..., J. Clin. Microbiol. 31, 
1293-1298, 1993): PvuII cleavage of traces of genomic DNA and subsequent 
amplification of IS6110 containing fragments. 
Upon request I will send you more detailed information on the advantages/
disadvantages associated with the individual strategies.

An NIH task force headed by Dr. Holtzmann at Johns Hopkins Sch of P.H. is
presently considering all of the issues of lab standards in genetic
testing.
They are also considering whose responsibility it should be to ensure that

Studies of the biochemistry of disease, coupled with advances in 
recombinant DNA technology have led to massive commercial interest in 
the production of highly purified, stable proteins. Apart from protein 
therapeutics, expanding markets also exist in diagnostics, industrial 
enzymes as biocatalysts, veterinary products and molecular biology 
reagents.

This protein biotechnology course addresses developments in protein 
technology from protein extraction, through the various protein 
purification stages, to their presentation as stable products. The 
respective merits of various methods for protein characterisation and 
quality control are compared, issues of protein stability are treated in 
detail, and topics of current interest, such as protein engineering and 
protein glycosylation are reviewed.

SDS (Sodium Dodecyl Sulfate), pH approx. 13 .  How does my friend keep

Currently I am trying to propagate bee picornavirus using tissue 
culture cells.  So far the wasp cell culture does not appear to be 
able to grow this virus.  Has anyone had any luck with this type of 
viurs in any other type of cell culture systems?

The following is the summary of a paper which has been accepted for
publication by J. Clin. Microbiol.

Of particular interest is the fact that the DNA probe  and  the PCR
protocol specific for Porphyromonas gingivalis were derived from the
sequence of a random amplified polymorphic DNA (RAPD) species-specific
fragment. We are currently developing  similar procedures for the diagnosis
of other fastidious gram negative anaerobes in mixed infections.

Immunomagnetic PCR and a DNA Probe for the Detection and Identification of
Porphyromonas gingivalis.

The aim of this study that we describe was to combine an immunomagnetic
separation and a polymerase chain reaction followed by dot blot
hybridization with a DNA probe for the detection and identification of
Porphyromonas gingivalis. Immunomagnetic particles were coated with
monoclonal antibody specific for P. gingivalis and incubated with a
suspension of seven oral bacterial species spiked with various dilutions of
P. gingivalis. Beads with their load of bound bacteria were boiled in
water, and the target DNA in the supernatant was amplified with a primer
pair to generate a 593-bp PCR fragment specific for P. gingivalis. Finally,
the product of amplification was detected by dot-blot hybridization with a
digoxigenin-labeled 593-bp probe. The detection limit was determined at 100
bacterial cells/ml. The immunomagnetic-PCR/DNA probe procedure described
here should be useful for the rapid, specific and sensitive detection and
identification of P. gingivalis in clinical samples.

Hi all...

I am curious on the advantages of spinner cultures or roller bottles over 
tissue culture flasks to propagate picorna viruses in vero cells.  For 
example, are the viral yields significantly greater compared to tissue 
culture flasks?  Also, can I use the adherent vero cells in a spinner or 
roller system?  Gee, any advice is greatly appreciated.... Thanks in

I believe that Roche Diagnostics (a division of the Swiss pharmaceutical giant Hoffman

Enterocytozoon bieneusi. I would be testing the feasibility of their use
