I can't address the potato aspect, but I ahve used chemiluminescence with
several animal viruses. In terms of sensitivity, I have found the light to be
quite sensitive. Existing WB protocols need to be modified.

For example using a similar anitgen/antibody system, when moving from I-125
protein A detection to chromogenic substrates, we had to dilute the primary
an additional 5 fold for comparable sensitivity. When we moved to chemi, we
diluted an additional 4 fold after again. Another pecularity I have found is
that the manufacturer's suggested dilutions of secondaries are never enough.
I recall that Amersham recommended 1:1000, but we dilute 1:20000 - 1:50000
for best results.

The trick to tell which is too much (which is usually the problem with these
more sensitive methods) is to discern the nature of the background. If too
many bands appear, dilute your primary; if the overall background is too
dark, dilute your secondary.

We are developing an assay for myeloperoxidase (MPO) in human granulocytes.

Current concept is to isolate WBC from whole blood using Pall Leucocyte 
filter.  Then lyse the WBC to release the MPO and use hydrogen peroxide as 
substrate, TMB as chromogen.

But Pall leucocyte filter is very very expensive.  Anybody has any experience 
with other material for this purpose?

While we haven't worked on viruses or used
Western blots, we did find chemiluminescence at
least as good as P32 in southerns for detecting
Erwinia carotovora subsp. atroseptica in potato.

aspirin company) to distribute its 1 step test for HIV in several asian 
countries where HIV is pandemic.

	    BIOSCI is about halfway to its funding goal!!

I'm interrupting the usual monthly posting of the BIOSCI miniFAQ to
bring you up to date on BIOSCI fundraising progress, a topic of
concern to your future use of this resource.  Thank you in advance for
taking the time to read this message carefully.

Last year we announced that BIOSCI was going to adopt the U.S. Public
Broadcasting System model to fund its operations after our DOE/NSF
grant runs out later this year.  Unlike PBS, we are not soliciting
contributions from users; we are only selling ads on our Web pages
solely to cover our operating costs.  Our goal is to seek sponsorships
until we build up an operating reserve of about $100,000 and then
cease further promotions until we need to build the reserve back up.
(The accountants among our readership will be familiar with the
problem of deferred revenue which we can not safely utilize until ads
have been displayed for a period of time.)  We are only about halfway
to our funding goal and need to raise further funds to avoid having to
curtail services at net.bio.net.  Fundraising is time-consuming,
however, and we need your help as explained further below.

Our operating costs consist of our network connection, phone lines,
hardware maintenance (we will be getting newer and faster hardware
soon!), plus 0.7 FTE of salaries covering UNIX systems admin,
technical support, quality assurance, i.e., testing, of our system,
and administrative costs (such as the time it takes to actually
find/write/call potential sponsors and raise money!).  Although the
BIOSCI staff does get compensated for a portion of the work that they
do, this project has always received a lot of free after-hours and
"vacation" time labor, so we hope that no one will begrudge the time
that we do charge to the project to serve you.  All of the three
part-time staff members, Dave Mack, Julie Lawrence, and myself, have
full time day jobs and families in addition to working hard to keep
this service running for all of you.  Julie and Dave Mack are
subcontractors for BIOSCI; my time that is charged to the project
defrays a portion of my regular salary instead of adding to my income.

Besides having to relocate the project, we were very busy this last
year building new infrastructure such as our WWW hypermail interface
to the system.  This was released last December along with scores of
WAIS indices for the newsgroups.  Virtually everything is complete,
although we do continue to find and fix bugs (many through your
helpful feedback!).  We are still having some problems with our WAIS
indexing.  The archives continue to grow rapidly.  We are running over
100 indexes now versus three previously and any systems crashes cause
greater havoc with the indexing than before!  We are still working to
fix this as fast as our resources permit and appreciate your patience,
but we have been able to automate a lot of the infrastructure to
reduce labor as compared to past requirements.

We have also implemented new software to make moderation of
BIOSCI/bionet newsgroups much easier and combat the growing problem of
Internet junk mail and USENET "spamming."  About 20% of our groups are
now moderated, many of them by the BIOSCI staff!  This, for example,
made a major difference last year in the quality of content in our
EMPLOYMENT/bionet.jobs.offered newsgroup which many commercial
concerns and recruiting firms are using **without charge** to recruit
candidates for positions in the biological sciences.

We are also now in a position to have sponsors for individual
newsgroups as you will have noticed if you have visited
http://www.bio.net/ and clicked on "Access the BIOSCI/bionet
newsgroups" recently.

So, how can you help??

As noted above it can take a lot of time to contact potential sponsors
if I have to do it all myself.  Our request is quite simple.  You can
do two important things which will take very little time for you
individually.

Your usage helps attract sponsors.  If you contact any of our
sponsors, please be sure to thank them for supporting BIOSCI.  It is
critical for them to get this feedback if they are to continue their

communities of scientists (and they haven't been heavily promoted
yet).  While these may be valuable niche markets to some advertisers,
it will generate more labor and overhead having to find these
sponsors, fairly price the locations, and deal with lots of smaller
sponsorships than fewer mid-to large sponsors.  We are striving to
keep our operation as lean and efficient as possible since we are not
trying to make careers out of running BIOSCI.  We are trying if at all
possible to avoid the administrative overhead entailed with processing
lots of small payments to reach our fundraising goals.

Dear colleagues,

I'm a specialist in Infectious Diseases in a 900 beds university hospital and I'm trying to make guidelines for residents in 
the Emergency Room to help them in managing infectious diseases.
I'm looking for criteria to decide when to perform blood cultures, blood analysis; when it is safe to give patients oral 
antibiotherapy and send them back home. I'm aware of the SCAPS score as well as guidelines when to hospitalise patients with 
pneumonia, but I'm seeking easier decision trees possibly efficient to manage most patients admitted for fever. Does somebody 
have any idea about these criteria, personal experience in guiding non infectious diseases specialist physicians in the 
treatment of infectious diseases, or where I could find some information about that topic ? Thanks in advance.

Can anyone give me information in regards to the new CLIA regulations for use of

A bit of a potato centric answer for you I'm afraid:

I know of one company in the UK that uses dot blots to detect potato 
viruses in tubers.  We (Scottish Agricultural Science Agency) use dot blots
 to detect potato spindle tuber viroid in potato plants received in our 
quarantine facility.  

PCR is routinely used in the EC for testing for Pseudomonas 
solanacearum (Brown Rot of potatoes) and Clavibacter michiganensis 
subsp. sepedonicus (Ring Rot of potatoes). 

There are also PCR-based tests for potato cyst nematode 
identification that are coming into routine use.

As I understand there are many people researching and using molecular 
diagnostics for plant pathogens around the world (specifically PCR

However, not many of these tests have been successfully integrated 
into commercial services.
Is this indeed the case, and does anybody know of molecular tests for

There are several suppliers of slides for IFA, but to my knowledge
there are not other immunoassays that are commercial. Even if some do
show, beware of false positives. Early results indicate much potential
crossreaction. I do know of some working on this, but it probably
won't be available for about a year or so.

                                  This is a question for biosensor 
researchers.
Over the past 20 -30 years or so, there has been substantial research into 
the development of sensors for organo-phosphates. Some of these sensors are 
based on the inhibition of cholinesterases.

 Does anyone know if this type of sensor has ever been commercialized?

I understand that the military in the USA was interested in this type of 
sensor for monitoring nerve gases in the field; were useful sensors ever 
developed?

Can anyone point me to useful sources of information, websites etc on this 
topic?

Update:  PYNG Technologies (PYT.v)-- Pyng has recently completed an intense
evaluation at the B.C. Paramedic's association.  The evaluation was all day in front
of 11 different evaluators.

Pyng's FAST1 device was rated on a scale of 1-10 in many different catagories.
The full summary results (7 pages) will be available thursday at their web site.
You may be interested to know that they scored an average of over 9 out of 10.

The meetings with the army have also just been completed with similar results.

My mother recently had an irregular PAP smear. She has had
several in the past months. They seem to be having trouble
reading the test. She read something about someone using lasers
and computers to read the tests and getting more reliable
results. Does anyone know where this is being done or where I
can get more info?

To my knowledge noone has ever succeeded in automating
this test although many have tried.

You probably need to seek an outstanding pathologist
to glean a good interp. for your mom.

The Genetics and IVF Institute in Fairfax, VA (near you),
MCV, Eastern Virginia School of Medicine in Norfolk, and
Hopkins in Baltimore all have excellent departments for
this purpose.

Avail yourself of their services with dispatch.

Where can we find more info on this device that uses

Hi all,
Does anyone know precisely how or wether issuance of a patent for a gene
sequence like BRCA1 (or p53, DCC etc. etc.) affects the ability of a hospital
clinical lab to obtain and use (and charge for) DNA sequence information for
patient care?

	These sensors actually do measure arterial p02 levels.  Enough O2
permeates from the arterial vessels to the surface of the skin to allow

Commercial products  have been on the market for at least 20 years.  Much
of the Premie measurements have been replaced with oximetry, which of
course does not measure arterial p02 but can infer it based on the
assumption that the heart/lung functions are normal - not always a safe
assumption.

The patent holder can exclude anyone from making, using, or selling the 
gene for any purpose for twenty years from the date the patent was 
filed.  The only exemption is use for sholarly research or research 
designed to improve the invention.  Use in patient care would require a 
license.  However, if BRCA1 proves useful for breast cancer diagnosis or 
monitoring, I suspect Millenium, or one of its licensees will produce 
and sell detection kits.

For the information of others in the newsgroup, the Entero-test kit is a
noninvasive device for collecting H. pylori bacteria from patients with
stomach ulcers.  This method is especially useful for monitoring the
effectiveness of antibiotic therapy.  The kit consists of a mesh string
connected to a gelatin capsule.  The patient swallows the capsule, then the
string is retrieved after a half hour or so.  The string is placed on either
culture agar or urease agar to detect the organisms.  New methods for
detecting the organisms retrieved on the string are under development.

I would greatly appreciate to hear of diagnostic labs who perform
hepatitis E PCR. Also I would be interested in manufacturers of such a
test. I am working for a medical diagnostic lab, and we get occasional
requests. I would be very interested in opinions on the clinical
usefulness of such a test and its complementary value (?) to
serological testing. Thank you very much indeed.

Does anybody offer PCR tests for hepatitis E? Any manufacturers for
such a test? I am working for a medical diagnostic lab and we get
occasional requests. I would be very interested to hear opinions on
clinical usefulness, reliability, sensitivity as well. Thank you very
