Does anybody know the half life time of free ribosomal DNA in a
neutral solution?
My idea is to detect in a quantitive way the amount of viable
moulds on basis of RNA. The idea is that RNA has a short half life
and therefor a RT PCR of freshly isolated total RNA could tell
something about the number of viable cells. However I am not sure
about the stability of ribosomal RNA.

Hi everybody,
I am looking for a protocol to extract fungal DNA from human nails or
skin
(i.e, the keratinized tissue). Does anybody know how to release the
fungal
cells and get rid of the keratin.

thanks

We have come across a curious phenomenon when making cocktails of
monoclonals. Two of our mab/Alk.phos conjugates which detect Potato
Virus A
pick up an isolate of this virus individually in ELISA. However when a
mixture of the two conjugates is used we cannot detect this isolate. Any
ideas of what is going on?

Hi

we regularly assay for the t(14;18) rtanslocation which involves the
bcl2 oncogene and the IgH gene. Our standard positive control is a PCR
product of approximately 230bp. recently we obtained a patient sample
which gave a product of over 1000 bp.
My question is: how big do they get? is it possible that a false
negative could be obtained for very large and possibly non amplifiable
products. Is there a diagnostic significance for very large rearranged
products?

thanks for your help

There are major and minor breakpoints at this locus.
The rearrangement involves slippage along some Alu
(repetitive) DNA in the heavy chain gene. You should
read the literature. There are also indications that
normal individuals contain a small percentage of cells
which rearrange. PNAS paper around 1994-95 if I remember
correctly. So you need to really, really know what you
are looking for at this locus before coming to any
conclusions.

Good luck.
