Hi there!

This is a follow up on an earlier message sent by Martin Parr, and the reply
of Alastair Hamilton.  Concerning those weak bands that appear only on the
autoradiographs; does anyone know if they can be trusted (segregation etc.),
or if they are just artifacts? Why are some bands weaker than others
anyway...?

Post doctoral position is available to investigate the molecular genetics of
plant-plant interactions. We are interested in defining the signaling
pathways that regulate how plants grow in the vicinity of other plants. The
goal of the post doctoral research is to assign biochemical and in vivo
functions to transcripts hypothesized as signal mediators between plants
(Matvienko et al., 2001 Plant J. 25(3) 1-15. More details of our research

The successful candidate will have extensive expertise in molecular biology,
yeast genetics and/or biochemistry. Experience in bioinformatics highly

Hi Thomas,
I think it's fairly inevitable that  PCR amplification of multiple
templates is going to amplify some fragments more efficiently than others.
Further, because this is PCR, the tiniest variations in amplification
efficiency during early cycles will translate into enormous differences in
the concentration of different amplicons by the end.
Of course a PCR explanation for fainter bands this doesn't rule out
artefacts, though it isn't something I have seen.
Obviously, to discriminate between background noise and faint bands it's
necessary to set a threshold.  Providing bands that are close to this
threshold amplify consistently then they should be trustworthy.   Life,
however, is not just or fair and I have seen published work that reports
considerable differences in the relative intensity of different AFLP bands
between duplicates, though, again, it's not something I have experienced.
I'd  be very keen to hear from anyone who has.

I could dig out some references on this if that would be helpful
