Subject: PCR from Mucous

Hello

I hope there are some people on this list that can help. I am
seeking advice on how to extract DNA from mucous and/or whether it is
possible to PCR directly from mucous. This is part of a forensic
project on abalone where often the only clue is slime. We have a
system that works well on all sorts of processed tissue but I am
looking for a protocol to extract DNA from mucous. I presume that a
CTAB, Phenol, Etoh type protocol would be the way to go.

Any suggestions will be welcome.

Hello,
        I am writing to inquire whether anyone has experience or
protocols for in situ hybridization or in situ PCR in marine
molluscs.  Are there any particular problems associated with the
increased ionic strength of seawater vs normal cell culture
media?  What about the intracellular milieu?
