I have a question regarding the right choice of vectors for phage
display.

I am planing to display a 60 amino-acid EGF-like domain (with an even
number of disulfides) as a fusion to the M13 gene III protein, using a 3
+ 3 system.

I have two vectors (the Stratagene Surfzap and Pharmacia pCANTAB-5E)
available from projects in which an unrelated protein and ScFv's were
displayed.

I was wondering if somebody had experience with either one of these two
vectors that would help me to decide which one to use for the display of
the relatively small EGF-like domain?

I have used the Surfzap vector in the past but for this project I would
have to introduce an additional affinity tag and amber stop codon myself
to get the option of recovering and purifying soluble protein in
suppressor strains. That would be another unknown as far as expression
is concerned. Using Surfzap would also require much longer oligos at the
N-terminus because of it's unique location of the cloning site. I would
like to avoid this to reduce errors caused by the oligo itself.

At the moment I am in favor of the pCANTB vector. I would like to use
the amber stop codon and E-tag for the purification of soluble
recombinant EGF-like domains without the need for subcloning. The down
side is that I have not seen applications of this vector that involve
proteins other than antibodies.(I have to admit that I have not looked
very hard)

POSTDOCTORAL FELLOW

We have a position starting June 1998 on an NIH training grant in the
area of hemostasis, thrombosis and inflammation, financed up to three 
years.  We seek an individual with experience in protein biochemistry 
and/or molecular and cell biology.  The research involves the 
interaction of protein ligands with blood or vessel wall cells.  The 
techniques involve the use of peptides, recombinant proteins, 
monoclonal antibodies and transfected cells to probe the requirements
and consequences of the interaction between the ligand and its receptor.

No teaching duties are required.  The Sol Sherry Thrombosis Research 
Center of Temple University has existed as a research department in 
the Medical Schoolfor 27 years.  A multidisciplinary group of 
investigators who collaborate intensively and an extensive collection 
of common equipment and facilities ensure a stimulating and 
productive environment.

1) Using the WWW to access the BIOSCI/bionet newsgroups.

As of 10 December 1995, all BIOSCI/bionet full newsgroups are
accessible through the World Wide Web (WWW) at URL http://www.bio.net.
One can read and reply publicly or privately to both recent postings
and archived messages through one's Web browser if it is configured
properly to send e-mail.  Each newsgroup is equipped with its own WAIS
index.  The main BIOSCI home page also has access to the BIO-JOURNALS
Table of Contents database WAIS index and the BIOSCI user address
database described in another item further below.


2) What to do about "spams," i.e., junk mail, ads, etc.
-------------------------------------------------------
BIOSCI is a set of parallel USENET newsgroups (the "bionet" groups),
mailing lists, and a hypermail archive at URL http://www.bio.net/.
The same postings are distributed on all media (except for a small
number of mailing-list-only groups at net.bio.net).  Unfortunately it
is becoming a despicable practice on the Internet (by a few people out
to make a fast buck) to do automated mass postings to thousands of
newsgroups and mailing lists.  These attempts to grab free advertising
are refered to as "spams" in the usual, somewhat boneheaded, net
terminology.  USENET is more susceptible to this practice, and many
spams originate on the USENET groups and then are passed on to the
mailing lists.  However, spammers also get lists of mailing addresses
and hit these too, so neither medium is immune.

What should you do personally if you get junk mail?
---------------------------------------------------
Just delete it and move on without reading it further.  Filing a
protest is becoming increasingly useless because spammers are often
disguising the addresses where the messages are sent from.  Unless you
really understand Internet mail systems, your attempt at protest by
sending replies to the message will often end up being sent to the
address of an innocent person that the spammer is victimizing.

What can BIOSCI/bionet do to protect its newsgroups?
----------------------------------------------------
The only solution currently available is to moderate the newsgroup.
If this newsgroup is already moderated, then you are in good shape.
Moderation protects the USENET distribution from about 95% of the
spams that are being sent to date and protects the mailing lists
completely.  Moderation means, however, that someone has to take the
time to review each message before it goes out.  We have set up
software here that simply allows the moderator to forward to an
address at net.bio.net messages that (s)he wishes to have distributed.
This takes no more time than that needed to read the message and pass
it on, say about 1 min. per message.

Most newsgroups currently have a discussion leader who is responsible
for their newsgroup.  The discussions leaders and their e-mail
addresses are listed in the BIOSCI Information Sheet which is
available on the Web at http://www.bio.net/.  If a newsgroup is being
hit with too many junk postings, please contact the discussion leader
for that group and see if there is interest in moderating the group.
Please do not assume that by simply posting a complaint to the
newsgroup itself, anyone on the BIOSCI staff will act on your
complaint.  With close to 100 newsgroups to run, the BIOSCI staff has
to rely on the discussion leaders of each newsgroup to report problems
directly to us at biosci-help@net.bio.net.

We will moderate any of our newsgroups if the discussion leader tells
us that the readership of the group wishes to do so and if a moderator
is willing to do the work.  For most BIOSCI/bionet groups, this
entails only a few minutes of work each day.

Moderating a newsgroup will resolve probably 95% of the junk postings
on the USENET distribution.  Unfortunately there are easy ways for
determined spammers to override the moderation mechanism on USENET,
but we can protect our e-mail subscribers from unwanted postings if
the newsgroup is moderated.  You can also access our newsgroups over
the WWW at URL http://www.bio.net.  While this Web interface will not
stop spammers from trying to post to the groups, this will give you
yet another way, besides using USENET news, to keep the junk out of
your personal mail files.  For those of you with local USENET news
systems, the Web interface will also give you faster access to new
newsgroups and recent postings.


3) Examples of subscribing and unsubscribing to the mailing lists.
------------------------------------------------------------------
PLEASE NOTE: The BIOSCI management does NOT act on
subscription/unsubscription requests that are posted improperly to the
newsgroups and mailing lists.  People who do this only bother everyone
on the lists to no avail.  Please be sure to follow the proper
procedures below.

Here I would like to give you some information on the 
XVII meeting of the Swiss Society of Neuropathology, 
which will take place in St. Moritz (one of the most 
famous Swiss skiing resorts) in March 14-18th, 1998. 
This venue sports a 30-year-old tradition of bringing 
together neuropathologists, neuro-clinicians, and 
neuroscientists interested in disease mechanisms. 
We specifically welcome the participation of "Aerzte im 
Praktikum", medical students and PhD students, who will

3. How to submit abstracts
IMPORTANT: Deadline for submission is November 30th, 
1997. 
- The language of the meeting is English. We will have 
both oral and poster sessions. Since we anticipate 
fierce competition for space in oral communications, 
oral format cannot be guaranteed in advance. If you 
submit an oral communication, you will be notified of 
acceptance before Dec. 31, 1997. In negative case, you 
will be given the option to present your work as a 
poster. At any rate, each participant can contribute 
only one oral communication (but an unlimited number 
of posters).
- Electronic submission of the abstract is imperative 
for acceptance and for inclusion in the abstract 
booklet. The preferred way to submit an abstract is to 
e-mail it as a fully formatted word processor file 
attachment to the following address:

The nearest airport is Zurich. Trains circulate between 
Zurich and St. Moritz hourly, and the trip Zurich-St. 
Moritz is approximately 3.5 hrs. The rail timetable can

What happens if you need to cancel your registration and 
cannot come to the meeting? 

If cancellation is timely, your registration and 
accommodation fees will be refunded with the exception of 
our bank expenses. 
For late cancellation and "no show", fees will incur. 

Therefore, the policy outlined below will be applied:

- Cancellation before 30.10.97: 95% refund or higher. 
  A total of max. 5% of your remission must be held to 
  cover our bank and credit card costs. In certain cases,

The project will use a combination of chemistry and protein biochemistry to
create artificial proteins to target certain cells to elicit an immune
response with application for AIDS and cancer vaccine biology. 

Applicants must have a Ph.D. in chemistry or biochemistry, with experience of
peptide synthesis. Experience of protein structure modification would be an
advantage, and in exceptional cases could compensate for lack of
peptide-synthesis experience. Applicants should have a proven track record of
high-quality work, attested by publications in major journals.
