     I've been looking at a culture of Candida tropicalis under a microscope
at (1000 x) because subcultures from the primary culture weren't growing
well. I'm seeing something I need help explaining. Inside most of the cells
is an object about the size of a cell nucleus that's bouncing and tumbling
around in the cell very rapidly. I haven't noticed this in other cultures of
the organism. Is this a common artifact of microscopy? Is it a symptom of
something wrong with my culture? Thanks in advance.

Graduate Fellowships in Fungal Biology

Fellowships are available for suitably qualified graduate students to work 
with selected members of the Biology, Biochemistry, and Plant Pathology and

Its a parasite, not a mutualistic symbiont.  I've only seen 
it parasitising pine species, but it may affect other kinds

Dear Fellow Biologists,

	I am a student at Swarthmore College, PA and am currently taking a MicoBio
seminar in which we are required to present a proposal which we put
together ourselves.  I have chosen to work on tuberculosis (Mycobacterium
Tuberculosis) and to take a closer look at what possible alternatives to
using antibiotics would be.  Currently, isoniazid, ethionamide, ethambutol,
pyrazidamide or a combination of any of these are being used to treat the
disease.  Because of incorrect or incomplete usage of these drugs, however,
resistance is developing quite rapidly.  Drug resistant strains of TB can
be found mostly in Africa and some parts of India.  
	My question to you is the following: Can you help me find some references
on the activation of Macrophages in which the Mycobacteria remain for an
extended period of time?  If so, what is the name of the protein or factor
that does so and where does it map to?  So far, I have been able to find
some info on IFN-gamma, but not on its gene location...if you know
something I don't, please drop me a short note over e-mail...thanks

With regard to Kambucha again.  I see it made the "National
Examiner" (Nov. 8, 1994, p. 40), a US based gossip weekly,
with the following headline, "Magic Mushroom Tea Fights Off
Wrinkles".  It is said there to contain usnic acid.  They
also have special dishes for growing the "thing".  You
wouldn't believe the other claims.
 
With regard to eliminating mushrooms - nobody had any
brilliant ideas, aside from picking all of them or cementing
over the backyard, neither of which was necessarily
brilliant.

Hi there.

Just a very amateur mycologist here with some questions.

The growth of certain species of mushrooms seems dependent on sunlight, or
at least an artificial light if cultivated in the lab.  I am wondering if
mushrooms are inherently phototrophic, like photosynthesizing plants, or
if some species just benefit more from exposure to light.

Will photo-responsive mushrooms grow more vigorously under stronger light
as opposed to weaker light?  Like some plants, will certain mushrooms elongate
less and grow thicker if exposed to strong light?  And finally, if this is
the case, what chemical actions are taking place within the fruiting body
of the mushroom to make it a different form and function from its parent
mycelia?

Hoping these questions aren't TOO vague....

Why on earth would anyone wan't to get rid of mushrooms? Refer these people for
counseling!

Aternatively, they could scatter a 1:1 mixture of copper sulphate and sodium
clorate in their yard at a rate of about 500 grams per square meter.  This 
should eliminate all mushrooms and give a yard a very uniform look.

It is really heartening to know so many people feel so
kindly of the lowly mushroom.  I am no small fan myself. 
So some of you have been berating me for asking how to get
rid of mushrooms.  It comes with the job.  Every once in a
while we are asked not to love our subjects but to find
ways to control them.
 
For years I have advised people to pick and remove by hand
mushrooms if they find them annoying.  Most give up and
learn to live with them.  Then there came the case of the
deadly Lepiota.  A small, rare species which came up in
abundance in one yard under a Norway Spruce.  In the yard
lived 2 children, a toddler and a crawler.  The toxins are
amatoxins, for which there are no reliable antidotes, and
the effects are delayed.  The parent was reasonably
disturbed about such species and picked and removed them. 
But more came up, and sometimes from small inconspicuous
buttons that were missed on first examination.  The area
was fenced off, the lower tree branches were trimmed back
to help dry the area up, the needles were raked and
removed.  The mushrooms moved out into the lawn during a
wetter period.  The parent was worried that even though
visible mushrooms were removed, one of the children might
find one lurking among the grass.  After following this
case for several weeks (now months) I really wondered if
there were not some remedy unknown to me which would save
the parents a lot of grief.
 
Yes, yes, there are more important things in life, but it
doesn't seem out of line to ask of a mycologist, "How does
one get rid of mushrooms?"  Entomologists are asked how to
get rid of insects.  Plant scientists are asked to get rid
of weeds.  Admittedly few ornithologists are asked how to
remove birds, but even they have stoop to do so around
airports and in big cities (feed not the pigeons).
 
So far the solutions have been: concrete slabs, hand
picking, psychological councilling, and modified Bordeaux
mixture.  This is entertaining.

Oh Well!  Brilliance is where you find it, I do suppose.
But if *ideas* is what you need.....hope the following helps;
just off the top of my head.  If you try any of them, do 
let us know....

As with most gardening, I would see two basic approaches, of 
which you have already rejected one: weeding.

The other is altering the environment in a way that supports
the plants you want, or in this case discourages those you
*don't* want, since the desireable plant (grass) will coexist
very happily and non-competatively with the un (mushrooms).

	I. Chemicals, that is, find a soil ammendment that the
	mushrooms find discouraging but is tolerated by the grass
		A. ph balance
			1. more acidic
			2. more basic
		B. "Fertilizers", that is things that would be
		considered desireable for grass
			1. nitrogen
			2. phosphorous
			3. trace elements
				a. zinc
				b. copper
				c. etc etc
		C. Pesticides
			1. regular broadleaf lawn weedkillers
			2. insecticides
			3. antiseptics, suitably dilute
				a. lysol
			4. organic agents
				a. garlic infusion 

	II. Mechanical
		A. Humidity
			1. water less (protect from rain)
			2. improved drainage
		B. Soil characteristics
			1. add sand
			2. add clay
			3. areation
			
	III. Ecological - find a colonizer 
		A. earthworms
		B. insects
			1. ants
			2. pill bugs
			3. earwigs
		C. snails/slugs
		D. imperfect fungus
		E. bacteria
		F. different grass variety

Much domestication and breeding work on C. nuda has been done at INRA 
Bordeaux, Station Champignons, B.P. 81, 33883 Villenave d'Ornon, FRANCE, 
by J. Guinberteau et al.  A forthcoming paper by P. Callac in CJB devotes 
a paragraph to this subject, including citations.  50-80 tons have been

admonition more often encourages children to
disobey their parents - at least once - as we
all are guilty of defying parental authority. 
 I assure parents that no mushroom is going
to jump out of the ground and attack you. You
just need not to ingest them. In this regard
does anyone know of any mushroom which
has topically poisoned a person handling
a species? I do not.

You may want to write to Dr. Ian K. Ross, Dept. of Biological Sciences, 
University of California, Santa Barbara, CA  93106.  His son, Keith, 
wrote a masters thesis in the Geology Dept. there, on etching of glass 
and sand crystals by fungal hyphae.  The year was probably 1988.  Good luck!

An ungraduate student in the Laboratory of Mycology of the Universidad de
Buenos Aires, Argentina, is working with xilofilous Basidiomycetes. She is
making culture descriptions of the isolates. She has found, among others,
three sps. of Agarics: Oudemansiella canarii, Agrocybe cylindrica and
Gymnopillus spectabilis.
She would like to know if these sps. have been already described in culture.
It will be very useful any information and/or copies of papers and or
references dealing with culture studies in Agaricales.
She has copies of Pantidou et al (1983). Desjardin (1990), Beatrice Senn-Irlet
(1994) by now.
I will be veru grateful who anyone who can help her.

M. Pease wrote an interesting response to S. Redhead about eliminating mushrooms
from lawns.  At least a kernal of a thought...elevated levels of nitrogen have
been shown to reduce fruiting of fungi, particularly mushrooms that are 
mycorrhizal so adding a high nitrogen fertilizer may be able to surpress 
fruiting.  In one study of hypogeous fungi that I read, applying high nitrogen
fertilizer surpressed fruiting for four years.  I can't give you a reference...
I'm at home and the paper is at my office at school.  But if you're interested,
I'll look it up for you.

winter months under pine. These two fungi are a source of proteins used

I've been finding Exophiala spinifera and Rhinocladiella atrovirens (a related 
black yeast) on field grown maize ears from diverse locations in the US. My
interest in them stems from the fact that these fungal isoalates metabolize a
mycotoxin, fumonisin,  that often occurs in maize grain that is infected with
Fusarium moniliforme. There  is probably an interesting ecological story there
somewhere.

There is a wealth of information in the literature. There is also a

	Most guides describe the red capped scaber stalk, Leccinum 
aurantiacum, as edible and choice.  I, however, have found this to be 
untrue for myself.  My father and I grilled a beautiful specimen, along 
with some freshly caught kokanee, in our camp after a great day of 
fishing.  Pop, in his wisdom and general practice of avoiding boletes (he 
had a bad run-in with boletus soup years past), elected not to try the 
"delectable" treat.  The sliced mushroom fried quite nicely, adopting its 
characteristic black and slimy texture.  It went down well with the fish, 
too, for about the first two hours, that is.  What followed was not 
something I ever want to repeat, I vomited every hour until dawn.  I 
concluded, perhaps prematurely, that the mushroom I ate had disagreed 
with me.  I have no desire to test my hypothesis further.  Pop and I 
speculated that we might have some heritable sensitivity to some 
boletus-specific chemical.  I do not want to test this hypothesis either, 
and think it best to avoid boletes in the future.
	Of course I may have misidentified the mushroom, but I think it 
unlikely.  L. aurantiacum has the red cap, black scabers on the stalk, 
and characteristically stains first burgundy-red then purplish.  Consider 
yourselves warned, I for one will stick with morels and shaggy manes for 
awhile.

To all the Fungi People out there.

I'm conducting a laboratory experiment in Biology.  I've attempted to
study the life cycle of Schizophyllum using dry basidiocarps.  I only got
as far as the cotton-ball stage.

Eventhough I have about 23 slides made up, I'm still having trouble
picking out the major structures which emphasize the stages I've made
slides from.

I would greatly appreciate if anyone can give me any clues on how I can
differentiate between germlings (other than not being branched), primary
mycellium, and secondary mycellium.

Ryan -- tried to send this to you personally to your regular address; the

to your poisoning by the Leccinum.....
	Dear Ryan -- You have my heartfelt sympathy -- and appreciation 
for sharing your tale of woe.  I suspect you are correct -- that you and 
your dad share a sensitivity.  However, more people need to be alerted 
that not all mushrooms are safe to eat for everyone.
	Incidentally, did you cook your mushroom thoroughly?  (You might 
have mentioned that but I access email via long-distance and don't want 
to take the time to check what you said).  We have found via the Oregon 
Toxicology committee that in many instances undercooking is the culprit.  
Mushrooms should generally be reduced by a considerable amount before 
eating.  One of the most consistent offenders is the Armillaria mellea 
complex -- we had an instance in Oregon where twelve different people ate 
from a very large collection of Armillaria.  Eleven thgoroughly cooked 
theirs and ate modest amounts.  The gonzo guy who just lightly sauteed 
his missed Thanksgiving dinner the next day because the sight of food was 
more than he could stand.  And we have had numerous cases where 
Armillarias have been thrown into sauces or gravies at the last moment 
after light sauteing, with unhappy results.

I received a few replies concerning autonomous replication in Neurospora.
Most detailed was from fons.debets*fungen.el.wau.nl who said that he had
compared transformation of N.c. with a hygromycin vector alone and in 
combination with an autonomous vector.  They observed NO increase in
transformation frequency in Nc, unlike the 10-100 fold increase they
observed with A. nidulans and some other fungi.  
This suggests that the Aspergillus element does not function in Nc,
any other direct experience?

Help needed!

My father, aged 63, suffers on a chronic pneumonia and
bronchitis for more than one and a half year.
The begin set a temporary collapse of the immunic system,
due to too much stress in business (no HIV!).
Now, someone will have detected a mycosis of
*Aspergillus Niger* as cause of this symptoma.
He tries since Sep 1994 to cure this, with
moderate effect.

Who is informed about Aspergillus Niger mycosis ?
Is this a probable indication ?
Can someone help us ?

              Molecular genetics of Phytophthora infestans 
                  University of California-Riverside


A postdoctoral position is available to study the molecular genetics of
mating type in Phytophthora infestans.  The position is available for 2-3
years, with a possibility of renewal.

P. infestans is a diploid, oomycete fungus with with two mating types
(sexual compatibility types).  Genetic analysis has shown that a single
locus determines mating type.  DNA markers flanking the locus have been
obtained which will be used to clone the locus by chromosome walking.

Projects available include the cloning and analysis of mating type loci,
the construction of mutants, and the identification of genes regulated
during mating.  These projects involve chromosome walking, genetic and
physical mapping, differential cDNA cloning, gene analysis, fungal
transformation, etc.  There may also be opportunities to participate in
other projects in the laboratory related to plant-pathogen interactions.

Motivated candidates experienced in molecular biology or genetics are

Greetings everyone!  I haven't followed this particular newsgroup much, but 
I'm going to start.  My apologies if I am replowing old ground.

Does anyone have any information (reprints, the names of current 
researchers, etc.) on the influence of mold spores on indoor air quality?  I 
attended a symposium in Prague last summer where they were saying that a 
complex of dust mites, mold spores, and toxic chemicals (released from 
furniture, paneling, etc.) contributes to the "sick building" phenomenon.  I 
often get phone calls regarding mold in houses, and now I think I would like 
to pursue this as a research topic.  Any ideas?  Regards....
