  Hi,
I'm a genetics major at Texas A&M.  The other day I attended a lecture
on genome sequencing.  The professor mentioned a technique called RAPD,
but did not go into detail on it.  I've searched around trying to find
out about it  but all I seem to find is technical information on it.  I
was wondering if anyone could explain this process to me in more
layman's terms?  Thanks,

RAPD IS A VARIATION OF THE CONVENTIONAL PCR AMPLIFICATION THAT USES A 
THERMAL ENZYME TO AMPLIFY SEGMENTS IN THE TARGET GENOME, THE CONVENTIONAL 
PCR USES A FORWARD AND REWARD PRIMERS NOT RANDOM. RAPD USES RANDOM PRIMERS, 
THIS MEAN THAT YOU WILL DO NOT KNOW THE SECUENCE OF THE RESULTING DNA 
FRAGMENT.

IF TWO RANDOM PRIMERS FIND EACH OTHER IN AN  OPPOSITE DNA STRAND AND IN 
AMPLIFICABLE DISTANCE THEY PRODUCE A FRAGMENT WHICH REPRESENTS A RANDOM LOCI

This is a misunderstanding. RAPD DOES detect the heterozygotes (Aa or +/-),
except that it is not possible to differentiate them from the homozygous
(AA or +/+) genotypes, since the band in question is amplified from both.
(Earlier people tried to sort them on the basis of band intensity, but this
approach did not work.)

For this reason both +/+ and +/- give you a band (dominant) and only -/-
doesn't (recessive). This is why RAPDs are called dominant markers, since
the heterozygotes could only be detected by crossing them with a recessive
homozygote (-/-) and then assaying their F1s. All F1s with the selected
band in the given RAPD assay would indicate a homozygote (+/+) parent,
while 50% would point to a heterozygote (+/-).

There are a few codominant RAPD described in the literature. They might be
caused by a short deletion or insertion in between two suitably located
primer binding sites. In that case the recessive allele appears in the
pattern and its connection to the dominant one could be observed. Most of
the RAPD markers (>95% or more) are however dominant as you noted correctly
in your message.
