[Arabidopsis] primary and secondary At cell wall tissue

Hi, colleagues,
 
I want to know how to get the primary and secondary cell wall tissue from at plants less than one month old? Should I grow At plants to flowering before harvesting the secondary cell wall tissue such as stem?
