AB To assess the potential for applying artificial neural 
   networks to the identification of Gram-negative rods, 
   four different networks were trained ,differing by the
   number and quality of strains in the training and test 
   sets. A network was trained using 3429 strains and 
   tested with 6859 strains identified 95.5% strains at the 
   species level and 97.7%  strains at the genus level. The 
   sensitivity was 95.5 and specificity 99.9%.
   It was concluded that artificial neural networks are 
   useful tools for phenotypic identification of 
   Gram-negative rods.

TI Digital Film as an Aid for Studying Live Protists

AB Some aspects of conventional (non-digital) and digital 
   video imaging are described. Digital film clips stored 
   in a multimedia database may represent a cost-
   effective solution for managing large collections of 
   video images, retrieving specific images from the 
   collection, editing images and using the images for 
   research, education, and demonstration purposes. 
   A worthwhile application of digital film is the study 
   of movement in protists. This is exemplified by 
   observations on the beat of the transverse flagellum 
   of the dinoflagellate Amphidinium carterae, 
   metachronal waves in the ciliate Ophryoglena, and 
   the behaviour of the contractile vacuole complex in 
   the cryptomonad flagellate   Chilomonas sp. Problems 
   arising while querying digital image databases on 
   taxonomically difficult protists might be reduced by 
   interfacing these databases with a purely 
   nomenclatural database allowing the user to navigate 
   through lists of nomenclatural and taxonomic 
   synonymies.

   Instability and their Application to Recombinant 
   Strains of Saccharomyces cerevisiae

AB We have developed a set of Microsoft¨ Excel 4 macros 
   for the analysis of the kinetics of segregational 
   plasmid instability. The custom  SOLVER macro 
   is used to solve non-linear equations and the error 
   of the constants so derived is determined by the 
   jackknife method. Jackknife analysis is especially 
   useful when working with statistically complex data 
   in situations where the error distribution on the 
   required parameters is undetermined, even though 
   the distribution on the experimental data is known.
   The Jackknife macro utilises three component parts 
   to determine values for the rate of plasmid 
   segregation (R) and the growth rate difference 
   between plasmid-containing and plasmid-free clones (X). 
  Initial values of  R and X are determined 
   using two linear regression methods describing two 
   opposing situations (X>>R) and (X< or = R). These 
   values are passed on to the non-linear regression 
   module which also carries out the jackknifing. Although 
   jackknifed non-linear regression is time consuming, 
   parameters obtained from jackknifed non-linear 
   regression are robust. The macros work on both PC 
   and Macintosh computers equipped with Microsoft 
   Excel  and can be easily modified to carry out 
   jackknifed non-linear regression on other data 
   sets and models.
   As an example, this method has been used to 
   determine the effect of the absence of endogenous 
   2 mu m-plasmid on the stability of a 2 mu m-based 
   vector (pJB502) inserted into  Saccharomyces 
   cerevisiae. Improvement in plasmid stability in 
   2 mu m-plasmid-free hosts was shown to be 
   completely due to decreased R even though 
   X was the dominant factor in plasmid loss.
