AB More than six million of AIDS cases have been reported 
   since the beginning of the HIV/AIDS pandemic. In addition, 
   there were 21.8 millions HIV-1 infected persons by June 
   1996. In spite of recent improvements in the therapeutics, 
   the development of a preventive vaccine is critical for the 
   control of AIDS. The aim of this review is to present a 
   profound view of one of the most attractive regions of the 
   viral envelope: the V3 loop. In V3, the principal neutral-
   izing domain of HIV-1 has been located, as well as epitopes 
   for helper and cytotoxic T cells. Because of its excep-
   tional features, it has been included in the vast majority 
   of the vaccine candidates in evaluation. The main findings 
   about the structure and polymorphism within this region are 
   summarized. Despite its variability, certain structural pa-
   rameters are conserved in order to permit the V3 loop to 
   mediate important functions in the viral infection. A gen-
   eral hypothesis about the role of V3 loop in the events of 
   viral entrance and tropism is advanced. Different studies 
   demonstrate the existence of cross-reactivity among V3 pep-
   tides. Some human monoclonal antibodies recognize conserved 
   epitopes in V3 from different clades and neutralize primary 
   isolates. However, the relevance of these aspects for vac-
   cine development is still uncertain. Finally, an overview 
   of the main vaccine strategies involving V3 immunogens is 
   presented.

AB Bactericidal antibodies against outer membrane proteins are 
   crucial to protect against Meningococcal Meningitis. The bac-
   tericidal monoclonal antibody (MAb) CB-Nm.2, specific for the class
   3 outer membrane protein of Neisseria meningitidis, was assayed in
   an enzyme linked immuno-sorbent assay (ELISA) with a panel of 86 N.
   meningitidis strains. Fifty six strains be- longing to seven
   serogroups: A, B, 29E, L, X, Y, Z, and five serotypes: 1, 4, 5, 12,
   13, reacted with CB-Nm.2. The porB genes coding four such proteins
   were cloned and sequenced, and the translated amino acid sequences
   were compared with five previously published sequences. Sequence
   alignment re- vealed a five amino acid region (S/T)VETG located in
   the main variable region (VR) VR1 which was present in all N.
   meningi- tidis strains recognized by CB-Nm.2 and not in the strains
   which were negative in ELISA. Two synthetic peptides were de-
   signed on the basis of the predicted antigenic determinant for
   strains B385 and H355. Mouse antiserum obtained against the
   synthetic peptides recognized Neisseria strains in whole cell
   Dot-blot, but synthetic peptides failed to react with the MAb. The
   results show that the (S/T)VETG region is pres- ent among different
   serotypes of N. meningitidis and it is probably involved in
   antigenic recognition by the bacteri- cidal MAb CB-Nm.2.

AB A double-blind, phase-I clinical trial was carried out in 
   healthy volunteers to know the penetrability of topically 
   applied epidermal growth factor (EGF). Twenty-four indi-
   viduals were randomly distributed in three groups. The 
   treated groups received EGF (10 or 100 =B5g/g) in 1 % silver 
   sulfadiazine cream. The control group received the latter 
   cream alone. The serum EGF concentration was measured by an 
   ELISA. Negligible penetrability was obtained despite a 10-
   fold higher concentration, compared to the therapeutic 
   dose, was used. This low penetrability contributes to the 
   safety of the product reducing the possibility of systemic 
   or distant adverse effects.

AB We describe a protein sequencer, built-in with commercially 
   available elements. It uses zero dead-volume membrane 
   valves and a cross-flow-reactor. Solvents and reagents path 
   lengths are minimal. As a consequence, the consumption of 
   reagents is low. Miniature valves are used to control vac-
   uum and inert gas supply. Sequencer and the chromatographic 
   system are controlled by an IBM-compatible personal com-
   puter. A software was developed that offers several opera-
   tional facilities, allows the acquisition of the absorbance 
   signal and the chromatograms processing. The system can 
   control either analogue or RS232 high performance liquid 
   chromatography pumps. Sample amounts around 10 pmol can be 
   sequenced with an initial yield of 57-74 %, a repetitive 
   yield of 94-98 % and low background signals in chroma-
   tograms (<1 pmol).

AB Practical achievements of cancer immunotherapy research are
   not impressive. Forty years after the experiments with 
   chemically induced sarcomas showing tumor specific rejec-
   tion antigens, we still do not have a molecular description 
   of these antigens and cancer immunotherapy in the clinical 
   setting is limited to the intralesional application of BCG 
   in melanoma and bladder carcinoma, and to the use of inter-
   feron in a handful of rare malignancies. The seminal idea of 
   the papers delivered at symposium The Impact of Emerging Para
   digms: Immunotherapy in the Nineties, was the exercise of 
   placing cancer immunotherapy inside the theoretical context 
   built by autoimmunity research. The symposium was held at the
   Centro de Inmunologia Molecular, Havana, August 5-10, 1996.
