AB Widely recognized as an important defensive mechanism 
   against viral infections, interferons (IFN), especially 
   when present in the body for prolonged periods of time, 
   may be occasionally associated to pathological conditions 
   such as autoimmune diseases or degenerative disorders. 
   More recently prolonged IFN activation has been found in 
   advanced HIV infection at titers commensurate with the 
   disease activity. This IFN, that is antigenically identi-
   cal to human alpha IFN but is unstable at low pH, is ac-
   tually a mixture of alpha IFN with other acid labile 
   cytokines, including gamma IFN, whose simultaneous action 
   gives rise to a synergistic activity that is not longer 
   observed after inactivation of IFN gamma. In vitro this 
   IFN type can be induced by HIV infected cells or by puri-
   fied HIV glycoprotein gp120. In fact, peripheral blood 
   mononuclear cells (PBMC) cultivated in the presence of 
   gp120 show enhanced production of IFN alpha and gamma and 
   of IL-6, TNF, IL-1, IL-10, and IL-8. Moreover, a signifi-
   cant activation of B cells has been observed, leading to 
   the enhancement of both IgG and IgM release. Furthermore 
   a significant proportion of gp120 stimulated PBMC undergo 
   apoptosis. While high concentrations of IFN alpha inhibit 
   HIV replication and reduce infectivity of HIV from chro-
   nically infected cells, IFN gamma determines enrichment 
   of adhesion molecules on HIV envelope, leading to wide-
   ning of HIV host spectrum and infection of cells lacking, 
   the major CD4 receptor and to facilitated cell mediated 
   transmission. These data argue for a possible pathogene-
   tic role of IFN, at least under certain conditions of HIV 
   infection.

AB The gene tab3 contains the DNA sequence for 15 ami-
   noacids (aa) of the V3 loop from six different isolates 
   of HIV-1, joined by the sequence AGGGA. This gene was 
   fused to a DNA fragment encoding the first 26 aa of the 
   human  interleukin 2 (hu-IL2) and cloned in an expression 
   plasmid. The fusion protein (TAB4) was expressed at high 
   levels in different Escherichia coli strains. The removal 
   of the hu-IL2 fragment from the plasmid abolished the ex-
   pression of the protein. Similar results were obtained 
   for plasmids pTAB7 and pTAB7SE, containing two copies of 
   tab3 with and without the hu-IL2 stabilising sequence, 
   respectively. Plasmids pVB1 and pVB2, in which the gene 
   tab3 was fused to the bacterial signal peptides OmpA and 
   PelB, were also evaluated. Proteins VB1 and VB2 were ex-
   pressed at low levels and were not translocated to the E. 
   coli periplasm. Our results indicate that the lack of ex-
   pression of proteins without hu-IL2 was neither a conse-
   quence of the stalling of ribosomes at rare AGA codons, 
   nor a product of a more complex secondary structure 
   around the Shine-Delgarno region or the AUG triplet. The 
   analysis of steady-state levels of specific transcripts 
   showed that differential availability of functional mes-
   senger ribonucleic acids determined these differences in 
   the expression of the multi-epitope polypeptides.

AB We studied the dermic and eye irritancy effect of hu-
   man recombinant epidermal growth factor. We used the 
   Draize test in albin rabbits to determine the dermic and 
   eye irritation indexes. We did not observe signs of skin 
   irritation and the value of the primary irritation index 
   was 0. The conjunctival effects were mild and disappeared 
   after 72 h. The eye irritancy index was less than 10. We 
   concluded that this formulation is not irritant and can 
   be used in therapy.

AB Monoclonal antibodies against luteinizing hormone (LH) 
   were produced immunizing BALB/c mice with LH isolated in 
   the INEN. They were IgG1 class and they were purified 
   from ascitis fluid by affinity chromatography using pro-
   tein A Sepharose. The antibodies were evaluated by the 
   isotopical method in liquid phase and two of them were 
   selected for a sandwich ELISA, according their charac-
   terization. This system was used for LH determination in 
   serum samples, between 0.7 and 100 mU/mL and the sensi-
   bility of 0.2 mU/mL.

AB Monoclonal antibodies (MAbs) are produced in mice by 
   the intraperitoneal injection of the MAb-producing hybri-
   doma cells. The laws for animal protection and the regu-
   lations required for the production of MAbs for human use 
   are more and more strict and have lead to develop new 
   techniques to obtain these molecules. In this paper we 
   compare a new bioreactor system: the cell culture auto-
   matic system (SACCEL) (ICID, Havana) with a well known 
   commercial system: ACUSYST-R (Endotronics, Minneappolis, 
   USA). Taking into account quality, subclass and immunopu-
   rification performance we did not obtain significant dif-
   ferences regarding the bulk harvest yield and MAb quality 
   when we used the MAb CB IFN 2.4. We reduced the medium 
   pump rate in relation to MAb production and found that 
   even a 50 % reduction of the culture medium consumption 
   did not affect the specific antibody production. This re-
   sult represents a 13 % reduction of the production cost 
   per unit.

AB During 1992-1993 Cuba was affected by an epidemic neu-
   ropathy (EN) classified as a new emerging disease. Toxic, 
   nutritional and biological factors were argued to be in-
   volved in the etiology of the disease. The toxic-
   nutritional hipothesis was supported by some laboratory 
   evidences and may acted as a predisposing cause for the 
   disease. In the search for biological agents, samples of 
   the cerebrospinal fluid from patients and controls were 
   analyzed and a slow cytopathic effect was detected in 
   93 % of the patient=92s sample. The characterization of the 
   virus indicated that it was a picornavirus and suggested 
   that it was probably a mutated enterovirus or a new uni-
   dentified virus. Preliminary experiments suggested that 
   the virus associated with the EN fulfilled the Koch=92s 
   postulates. Our hypothesis is that the etiology of the EN 
   could be related with nutritional deficits and stress 
   that made the population more susceptible to the appear-
   ance of clinical symptoms because of the infection with a 
   virus, responsible for the neurological injury through a 
   probable autoimmune reaction against structures of the 
   nervous system, that otherwise, would be asymptomatic. 
   This paper is not intended to assay our hypothesis but 
   rather to summarize our preliminary results and to pro-
   mote scientific debate.

AB  In type 1 Gaucher disease, decreased activity of  
    glucocerebrosidase results in accumulation of
    glucosylceramide in macrophages. Infiltration of liver,
    spleen and bone marrow by lipid-laden macrophages leads to
    hepatosplenomegaly, bone lesions and cytopenia. These
    abnormal macrophages may produce and release macrophage
    derived factors and cytokines, which could contribute to the
    pathophysiology of the disease. Whether these cytokines and
    factors are elevated in Gaucher disease is currently unknown.
    In 29 type 1 Gaucher disease patients we measured serum
    levels of the macrophage derived cytokines IL8, IL6,TNFalpha,
    M-CSF and the monocyte/macrophage activation marker  sCD14.
    These factors were studied in relation to disease severity 
    and during treatment with enzyme supplementation therapy.
    Most patients showed remarkably elevated levels of  M-CSF 
    (2-8 fold)  and sCD14 (2-5 fold) as compared to normal
    controls. Levels of IL8 were elevated in all patients (2-20
    fold), whereas levels of IL6 and TNFalpha were normal. There was
    a significant correlation between severity of the disease as
    determined by the severity score index (SSI), and M-CSF,
    sCD14 and IL8 levels. M-CSF and sCD14 levels also correlated
    with the excess liver and spleen volumes. During treatment
    with alglucerase, levels of M-CSF and sCD14 declined, but IL8
    remained unchanged. The relative reduction in excess liver
    and spleen volume did not correlate with the relative
    reduction in M-CSF or sCD14 levels. We conclude that serum
    levels of M-CSF, sCD14 and IL8 are increased in type 1
    Gaucher disease. The biological activities of M-CSF and IL8
    may add to the pathophysiology of the disease.

AB  We present here the case of a Japanese female patient with
    aplastic anemia who developed monosomy 7 and clonal evolution
    following a treatment with recombinant human granulocyte
    colony-stimulating factor (rhG-CSF).  At the onset of
    aplastic anemia, cytogenetic analysis was 46, XX and X-
    inactivation/methylation analysis revealed a polyclonal
    pattern.  After 4 months of administration of rhG-CSF, she
    had 45, XX, -7 and a clonal pattern, although there were no
    morphological evidence of a myelodysplastic syndrome or
    leukemia. The ratio of monosomy 7 to normal analyzed by
    fluorescence in situ hybridization decreased after
    discontinuation of rhG-CSF and there were still no dysplastic
    changes and/or increased numbers of blasts.  These results
    indicate that the acquisition of monosomy 7 following rhG-CSF
    treatment dose not always cause clonal evolution to induce
    hematological malignancies.
