AB The epidermal growth factor (EGF) is a potent mitogen 
   for a variety of epithelial and mesenchyma derived 
   cells. Cells treated with EGF exhibit transient 
   metabolic changes and a morphologic phenotype which 
   mirror those found in malignant counterparts. EGF binds 
   to a specific membrane receptor with intrinsic tyrosine 
   kinase activity coded by a cellular proto-oncogene, 
   whose mutated form has been largely implicated in 
   malignancy. Experimental evidence indicates that EGF 
   potentiates chemical carcinogenesis in vivo as well as 
   viral transformation in vitro. Thus EGF has been 
   historically considered as a tumor promoter. 
   Nevertheless, recent experimental findings, mostly 
   derived from preclinical models, strongly suggest that 
   EGF treatment is insufficient to initiate benign or 
   malignant transformation. On the contrary, in certain 
   scenarios EGF may act as a potent cytoprotective agent 
   for normal cells, and sensitizes cancer cells to anti-
   tumor therapy. All these experimental data provide 
   fundamental basis for the clinical use of EGF in terms 
   of safety. The clinical experience accumulated SO far 
   suggests that EGF does not seem to initiate 
   carcinogenesis.

   peroxidase) with specificity confirmation. The IFN 
   alpha-2b antiviral activity neutralization was alSO 
   investigated. No anti-IFN antibodies were found by 
   ELISA in 184 not treated individuals while among other 
   samples the bioassay detected neutralizing activity in 
   subjects with viral chronic hepatitis (8/181; 4.4 %) 
   and neoplasia (11/121; 9.1 %). Ninety-eight healthy 
   subjects or patients with other illness were negative. 
   There was no positivity in leukocyte IFN-treated 
   individuals, 27 of them evaluated by ELISA and 52 by 
   the bioassay. After the treatment with IFN alpha-2b 
   positivity by ELISA was confirmed in a girl with 
   lymphangioma and in an adult with laryngeal 
   papillomatosis among 177 patients. These ELISA-positive 
   sera had neutralizing titers of 1:25 and 1:64 000 
   respectively. IFN alpha-2b treatment induced 
   neutralizing activity in 13.1 % (19/145) of the 
   patients evaluated by bioassay. The total incidence of 
   treatment-induced antibodies, evaluated by either 
   method, was 6.4 % (19/296). This is similar to what has 
   been reported for IFN alpha-2b.

AB Appropriate technology to enhance shoot induction 
   ability and vitroplants quality, characteristics 
   determined by the genotype but strongly affected by the 
   environment, might facilitate the generation of 
   transgenic crops. 1. In the tomato variety Contessa 98 
   % of the leaf explants formed callus but shoot 
   induction: a) was observed in 59.97 % of the explants 
   being 2.4 the mean number of shoots per explant if the 
   back side of the leaf disks were placed in contact with 
   the medium. Half as much was the percentage obtained 
   putting the other side of the explants in contact with 
   the medium; b) was modified by age and period of the 
   year in which the leaf tissue of the donor plant were 
   developed since 95.7, 90.2 and 49.6 % of the explants 
   obtained from 4, 6 and 8 weeks old plants respectively 
   developed shoots being, in the same order, 4.27, 4.32 
   and 2.61 the mean number of shoots obtained per 
   explant; c) was detected in 98 % of vitroplantlet leaf 
   tissue explants and 3.27 mean shoots per explant were 
   obtained. 2. Similar performance of the plants obtained 
   from seed and from leaf tissue was observed, in 
   particular fruit quality and yields, when all of them 
   were cultured under soil conditions.

AB The epithelial response to a daily topical treatment 
   regime with three different epidermal growth factor 
   (EGF) concentrations was evaluated in five pigs. 
   Animals were kept under controlled environmental 
   conditions, and 30 partial-thickness wounds were 
   inflicted on the dorsum. These were assigned to 
   treatments with 50, 10 and 2 µg of human reacombinant 
   EGF (hr-EGF/g). Control wounds received cream vehicle 
   or were untreated. Wounds were cleansed, inspected and 
   treated every day accordingly. The wound-derived 
   exudate was collected from the untreated wounds during 
   the first 24 h post-injury, and filter-sterilized to 
   qualitatively estimate the presence of protease 
   activity on a fluorescent synthetic peptide by 
   electrophoresis. After 7 EGF treatments, pigs were 
   sacrificed on day 8 and the wounds were harvested and 
   processed to determine the percentage of total re-
   epithelialization, average radius, and wound perimeter, 
   as the linear ingrowth. The computerized morphometric 
   study proved that only the largest dose significantly 
   stimulated re-epithelialization, whereas the other 
   doses were ineffective. It was alSO shown that the 
   acute phase-wounds exudate exhibits proteolytic 
   activity which completely degraded the fluorescent 
   substrate. This proteolytic activity was similarly 
   inhibited by 0.5 and 5 µg of a Kunitz-type protease 
   inhibitor. Our results lead us to speculate that the 
   bioavailability of topically applied EGF to accelerate 
   wound healing may be reduced by proteases derived from 
   local acute wounds.

AB Since the human immunodeficiency virus (HIV) was 
   proposed as the cause of the acquired immune deficiency 
   syndrome (AIDS), many investigations have been 
   conducted to establish the relationship between these 
   two entities. Nevertheless, as alSO described for other 
   viral diseases, the validity of the Koch's postulates 
   has not been established for the HIV. Although most of 
   the researchers consider that the HIV is the cause of 
   AIDS, few argue that no relationship exists between 
   these two entities. AIDS is characterized in most of 
   the infected people by a debilitation of the immune 
   system, followed by infectious and non-infectious 
   diseases which finally result in patients death. Taking 
   advantage of the exceptionally good conditions found in 
   Cuba for the epidemiology and control of the HIV 
   infection, we studied the incidence of tuberculosis 
   (TB), Kaposi's sarcoma (KS) and epidemic neuropathy 
   (EN) in HIV- (N = 11 004 563) and HIV+ (N = 1 303) 
   individuals between 1986 and 1996. The odd ratio was 
   calculated to compare the incidence of these diseases 
   in HIV infected and non-infected people. The results 
   indicated that for the diseases included in the AIDS 
   (infectious: TB, odd > 22 and non-infectious: KS, odd > 
   3100), the incidence was higher among HIV infected 
   individuals. For the NE, not included in the AIDS, the 
   incidence in HIV non-infected individuals was higher 
   than in HIV+ (odd = 0.49). These results added new 
   evidences showing that for our study conditions, a 
   cause-effect relationship exists between the HIV 
   infection and the AIDS infectious (TB) and non-
   infectious (KS) diseases.

AB Extraction and quantitation studies of the active 
   ingredient from an hydrophilic (potassium 
   estearate)cream prepared either with recombinant or 
   leukocyte human interferons, were done. The cream 
   samples were treated with different extraction 
   solutions including acids, alkali, surfactants and 
   organic solvents. Recombinant alpha interferon (IFN) 
   was best extracted using acetone (1-5 %) or using a two 
   phase extraction system composed by diethyl ether: H2O 
   (1:1). With this solutions about 80 % of the active 
   ingredient was recovered from the cream. In the case of 
   leukocyte IFN, the two phase extraction system using 
   diethyl ether: H2O (1:1) extracted up to 95 % of the 
   IFN. In addition other cream bases were assayed for the 
   formulation of the IFNs creams. High recovery (99 %) of 
   the active ingredient was obtained using diethyl ether: 
   H2O (1:1) for the following creams: a) Potasium 
   estearate, b) base composed of polysorbate 80 (0.83 %), 
   propylenglycol (10 %), cetilyc alcohol (1.14 %), 
   glyceride monoestearate (6.67 %), methylparabene (0.18 
   %), propylparabene (0.02 %), H2O: csp, combined with 
   carbopol 940 gel to 0.5 %, and c) the same as b) 
   without carbopol 940.

AB The World Health Organization (WHO) Requirements for 
   the whole-cell pertussis vaccine describe the potency 
   test in a very general manner because it varies from 
   one laboratory to another; one of the reasons for that 
   variation could be the mouse strain used, therefore 
   every laboratory shall evaluate the best conditions for 
   its performance. To be considered valid, the effective 
   dose 50 % (ED50) value shall be between the largest and 
   the smallest immunizing doses and the regression do not 
   show significant deviations from linearity. The optimal 
   dose range is experimentally determined in each 
   laboratory. For the establishment of this assay in our 
   laboratory the OF-1 mouse strain was used. Different 
   immunizing doses were inoculated to get a surviving 
   response which had to fulfil a linear regression 
   analysis for be considered appropriate. Ten assays were 
   performed following the same methodology. Six resulted 
   valid. The logarithmic transformation of the doses used 
   and their survival percent showed a good linear 
   regression (R ? 0.99) in the range of immunizing doses 
   containing 2; 0.4 and 0.08 International Units (IU). 
   Eight lots of Diphtheria-Tetanus-Pertussis (DTP) 
   vaccine were alSO evaluated to prove that the potency 
   test worked well. All the assays were valid according 
   to WHO requirements. The methodology described in the 
   present paper was considered appropriate to evaluate in 
   our laboratory the potency of the whole-cell pertussis 
   component in DTP vaccines.
