Dear colleagues,

does anyone have experience/a protocoll for SEMing planktonic larvae
that have been fixed in Steedman's fixative and then transferred to
Steedman's preservative? Into which percentage of ethanol do I transfer
them as a start for the graded ethanol series folowed by critical point
drying without doing any harm (shrinkage) to them. I have checked some
literature and googled but couldn't find anything.

All hints welcome!

Happy Easter!

Dieter

Steedman’s Preservative, Johnson and Allen (2005)
1. 95% water, RO or tap water ok
2. 4.5% 1,2 propanediol (propylene glycol)
3. 0.5% propylene phenoxytol

You can make a concentrate as follows:
90 mL propylene glycol
10 mL propylene phenoxytol

Add 5% of the concentrate to 95% water.

Steedman’s Fixative
1. 50% Formalin (40% formaldehyde) buffered with borax (20-30 g to 1
liter of full strength formalin)
2. 45% 1,2 propanediol
3. 5% propylene phenoxytol
