There are two other reports on using small amounts of plant material in PCR
reactions:
U. Langridge, M. Schwall and P. Langridge. 1991. Squashes of Plant Tissue as
Substrate for PCR. Nucleic Acids Res 19:6954
P Langridge, Univ Adelaide, Waite Agr Res Inst, Ctr Cereal Biotechnol, Glen
Osmond, SA 5064, Australia.
P. Berthomieu and C. Meyer. 1991. Direct Amplification of Plant Genomic DNA
from Leaf and Root Pieces Using PCR. Plant Mol Biol 17:555-557
C Meyer, Inra, Biol Cellulaire Lab, Route St Cyr, F-78026 Versailles, France.
I hope this also helps.
