Which Agrobacterium strain?

Dear All,
          Here are the replies that I got form my query as to which 
Agrobacterium strain was best for Arabidopsis transformation.  Thanks to all

Marc-
It depends on what type of transformation procedure you are using. LBA4404 
is a wimpy strain that can be killed by vancomycin and therefore is great
for root explant transformation.  However, the robust nature of the C58 
strains probably makes them better for in planta transformations.

I have used LBA4404 to transform Arabidopsis without problems. For some time my
lab has been using EHA101 (kan resistant), a disarmed derivative of A281,
because it grows faster than LBA4404. My binary vector has GentR, if you use a
KanR binary you should use EHA105 (kan sensitive). C58 is a wild type strain and
therefore not suitable for transformation and regeneration of normal plants.

Marc,
There are some reports indicating that nopaline strains of Agrobacterium are
better than octopine.  C58 is a nopaline strain, and LBA4404 is an ocotpine
strain.  However, we have been using LBA4404 for some time, and it is
working very well with the "Wo" ecotype of Arabidopsis.  This brings up
another consideration, ecotype.  LBA4404 would not work on Landsberg erecta.
We are interested in this and actually looking at in more detail.
Comparisions of octopine vs nopaline strains with the same binary plasmid vs
ecotype. Clearly there is an interaction.  We really dont have any good data
at the moment.

transformation of A. thaliana: comparison of the efficiencies
with various organs, plant ecotypes and Agrobacterium strains"
for a detailed comparison of some various possibilities.

Dear Marc,

    I guess I'm in the minority, but in my hands LBA4404 has worked remarkably well.  Perhaps the LBA4404 most people use has "gone bad".  The strain from Clontech (Palo Alto, CA) works great.
