This is a rather interesting observation. The fact that there is a stopcodon
in the MCS of pBI101.3 can be deduced from the information given by Clontech
in their catalog, BUT they also state that "GUS gene fusions either
transcriptional or translational, can be constructed by inserting fragments
in the MCS." This is misleading since some sites CANNOT be used for this
purpose and a MCS is supposed to give one some flexibility in cloning strategy.
So I feel very sympathetic with the reported problem and think Contech should
adjust the information given in their catalogue.
