DAPI Staining

I am looking for a procedure to stain Arabidopsis endosperm nuclei, with
DAPI  or other  flourescent dye ( that would not decay quickly). I  would
like to use this  to quantitate DNA amounts and estimate ploidy  in these
nuclei.
