Hi, this is a late response to Rishi's question for nuclei isolation method. We have published a simple nuclei-based method for preparation of mega-base plant DNA (Nucl. Acids Res.1994,. 22, 2169-2169).With this method nuclei can be isolated from both fresh and frozen tissue (whole plants or leaves). Yao-Guang Liu
