phase partitioning

Dear netters

I am currently trying to purify plasmamembranes of whole Arabidopsis
plants. I use the two phase-partitioning method, but only with partial
success.
Therefore,  I really would like to know about your experiences with this
method. Every input on this topic will be welcome. Which parts of the
plants are you using?
