Dear Netters,
I have been using the "floral dip" method for Arabidopsis transformation
by Agrobacterium-infiltration without vacuum since the first rumors
about it appeared in this newsgroup, but never found a literature
reference for it although the method is widely used.
Now there is a literature reference describing it:
Bent, A. F., and S. J. Clough. "Agrobacterium germ-line transformation:
Transformation of Arabidopsis without tissue culture." In: Plant
Molecular Biology Manual (Second Edition), S. B. Gelvin and R. A.
Schilperoort, eds. (Kluwer Academic Pub., Dordrecht, The Netherlands).
1998, section B7, pages 1-14
The authors also provide a short protocol on their homepage:
http://www.cropsci.uiuc.edu/~a-bent/protocol.html
Infiltration without vacuum really saves time and effort, and the
transformation efficiencies in my hands are very similar with both
methods.
