I am looking into doing transient expression in arabidopsis (and tobacco) protoplasts. My expression plasmid is 3.5 kb. Would I get away with using an insert gene of 2.7 kb, making a total final plasmid size of approx. 6.25 kb, or is this too large for efficient transformation? Thanks, Steven Spencer ---
