he
Arabidopsis thaliana  =DF-glucosidase and =DF-galactosidase genes and/or
to investigate the mechanism of substrate specificity in
=DF-O-glucosidases  and myrosinases (=DF-S-glucosidases).

LOCATION: Department of Biology, Virginia Tech, Blacksburg, Virginia
USA 24061-0406.

DURATION: 2-3 years, starting January - February 2002.

RESPONSIBILITIES:
Construction, cloning and expression of =DF-glucosidase and
=DF-galactosidase cDNAs in bacteria, yeast or any other suitable host
system; purification and biochemical characterization of
=DF-glucosidase and =DF-galactosidase isozymes encoded by cloned cDNAs
with special emphasis on substrate specificity; and isolation and
characterization of the natural substrates hydrolyzed by each
isozyme. There will be also opportunity to learn homology modeling
and protein crystallization for 3-D structure determination.

QUALIFICATIONS:
The successful candidates for the position should have a strong
background and publications from previous work in molecular biology
or protein biochemistry.  Experience is necessary in standard
recombinant DNA techniques including cDNA synthesis, cloning and
expression, site-directed mutagenesis, and enzyme purification and
characterization.  Experience with plant systems is not necessary.
Candidates who have prior experience with protein expression in
yeast, E. coli and or baculovirus systems will be given preference.
