I am trying to isolate BAC DNA in large quantity. The intention is to perform restriction digest of the BAC DNA for the eventual subcloning and invivo complementation. I have been remarkably unsucessful in purifying any significant quantity of DNA. Does anyone have a good protocol or other useful suggestion. ---
