I am a 5 th year biology student and I am trying to designe primers for PCR reaction using the Primer3 software. Could anymore tell me if it matters which strand I designe the primers? If I subbmit the 3'-5' sequence to the program (eventhough it asks for 5'-3' sequence)will the primers still amplify the desired fragment of DNA? Than you Marta ---
