There is a Shareware one called 'counter' , which is not to specfic.
I can send it to you by EMail.

It will let you specify the areas of a dig. image, and will
count the intensity of the grey-scale of the specified area.

Maybe if you have only black/white this will be what you want.

Great program, simple to handle, MS-DOS / WIN3.1  needed.

No experience, but I will try this (if I get a CCD camera).
