Assuming you mean experimental measurement rather than some kind of
computerised prediction procedure, there are several possibilities: 

Spectroscopy (UV, IR and CD)
Light-scattering
Chromatography
Dye-binding
NMR
X-ray and neutron diffraction

to name a few approaches used for biological macromolecules. It might
help if you were more specific about what sort of molecules you want to
work with.
