Help Tetrahymena Mating

I'm having trouble getting my Tets to mate.  Grown in Neff's at 28*C 125RPM.  S
tarved O/N in 10mM Tris @ 7.4  @28* with shaking 125 RPM.  Mated @ 1*10^5 in pe
tri plates.  Pairing is 80-90%.  only 5% or so complete conjugation and go on t
o express 6MP resistance (427.2 X428.1) Using millipore water and dedicated gla
ssware
