TNT rabbit reticulocyte

Has anyone out there used Promegas TNT rabbit reticulocyte lyste system for in 
vitro production of proteins?

I have used it for a 53kDa protein and have found no distinguishable 
differences between the control luciferase and test lanes when examined by 
autoradiography.

The plasmid containing the gene of interest is pcDNA3 (Invitrogen). Can anyone 
help!
