I isolate lymphocytes from human blood using gradient centrifugation method.I put the isolated lymphoctes in the RPMI1640 plus 10% FBS in the incubator. I found about 20-30% of the lymphocytes their shape changed to irregular from initial spherical shape. Would you suugest any methods and medium for good maintainence of the lymphocytes in the incubator ?
