NEED HELP WITH OSTEOBLAST ISOLATION!!!

Hello all you Cell Biologists,

I am currently involved in a research project that involes isolation of 
OSTEOBLAST from rats.  The technique that I am using for EXPLANT and
DISASSOCIATION is not working for me.  The media that I use is 2,3mM MgSO4
DMEM w/ 1% P/S & 15% C/S.
