Does anyone have a good procedure for preparation and storage of HeLa cell extracts for use in SV40 DNA replication. We have tried the method of Li and Kelly and that of Wobbe and Hurwitz. The initial activities are sometimes OK but are rapidly lost when stored at -70C.
