Jocelyne Yep no problems with that ligation - its a bog-standard mol. biol method. Do the ligation at 12oC, overnight, as the blunt end ligation is less efficient. Also, most of the rapid ligation kits supplied by the major manafacturers seem to work well in others hands in our lab. Use a higher, 10:1, ratio of insert:vector molecules.
