1. Study on correlation between cytoskeletal filaments and denuceation
process
in erythroblasts.
---Examining the dynamic change of quantitative and spatial distribution of
cytoskeletal filaments (vimentin, tubulin and actin) in different
developmental
stages of cultured splenic erythroblasts infected with anemia-inducing
strain
of Freind virus (FVA cell) using immunofluorecence technique and laser
confocal
scanning microscopy.
2. Study on apoptosis in FVA cells.
---Observing the change of cytoskeletal filaments and morphology.
---Determining the intracellular calcium concentration.
---Detecting DNA cleavage with in situ nick translation and
electrophoresis.
3. Subtractive cloning of gene related to erythroid differentiation and
denucleation in FVA cells.
---Isolating and culturing the splenic erythroblasts.
---Extracting and purifying mRNAs in two different developmental stages.
---Constructing two cDNA libraries.
---Identifying cDNA fragments related to erythroid differentiation and
denucleation using subtractive hybridization and PCR. Confirming by using
Northern blot assay. Four differentially expressed cDNA fragments are
cloned
and sequenced. One of  them has been submitted to GenBank (AA114369).
---Demonstrating the specific expression of the genes with in situ
hybridization.
---Subcloning the differentially expressed cDNA into pLXSN retrovirus
vector
and transfecting it  into MEL cells.
---Observing the cell phenotype alteration of MEL cells in growth rate,
cell
division index, colony forming rate and expression of beta-globin.
4. Cloning and expression of IL-6 in BL21(DE3)/PET-28 system.
