use of slo for cell permeabilisation

I have been trying to use SLO to selectively introduce pores in the 
plasma membrane of cells. I do this by incubating the cells with SLO  in 
an ice-water bath and then incubate the cells at 37 after washing the 
unbound SLO. However I find that the ER compartment is not really 
intact as I get a substantial amount of leakage. Can someone who has 
this system going give me some tips.
