Fluorescent cell number assay inquiry

Dear Cell Biologists,
I want to use a microtiter format for cell number determination assays
using a nucleic acid-binding fluorescent dye (such as Hoechst 33258). What
is the best method to permeablize the cells to allow the dye to access
nucleic acids? Is Hoechst 33258 the best choice of dye? Do I need to remove
the culture medium, or can I just add a detergent and dye mixture? Can
anyone recommend a good general reference that discusses these issues?
