Dear Cindy Tong,
The most common fixative uesd is Carnoy's fixative which is basically 3parts methanol and 1 part acetic acid.
Your cells have to be thoroughly washed in fixative before proceeding to staining.
If you are only required to do a chromosome count without any banding then Giemsa stain is sufficient.
