I've never done it myself, but a quick PubMed search revealed the
following:

Comparision of bilirubin conjugation in encapsulated hepatocytes,
hepatocyte homogenate and intact hepatocytes.  Bruni S, Chang TM

2.  Hepatology 1981 Nov-Dec;1(6):622-7 
Bilirubin mono- and diglucuronide formation by human liver in vitro:
assay by high-pressure liquid chromatography.  Chowdhury JR, Chowdhury
NR, Wu G, Shouval R, Arias IM

They both used HPLC.

The following is too old to have a PubMed abstract, but sounds
potentially useful:

3.  Anal Biochem 1972 Dec;50(2):652-5 
Determination of bilirubin in liver homogenate with diazotized
p-iodoaniline. Improvement of color recovery by incorporation of
antioxidant (2,6-di-tert-butyl-p-cresol) in the reaction system.
