Pertussis toxin protocol needed badly.

Hi Everybody.

I am researching in a receptor that mediates its action via Gi-proteins in
frog skin. My problem is that I can not load the tissue at all. I have tried
to incubate with 1ug/ml for 14 hours with no luck.
Has someone got any experience in this?
The tissue is about 4 celllayers thick and it has a cornified layer toward
the outside. The cells that I want to load are situated just under the
cornified layer.
